Prevalence and mechanisms of non-target site herbicide resistance in important weeds of the Czech Republic

Abstract:

Over the past two decades, herbicide-resistant weed biotypes have increased in frequency and diversity, posing a significant threat to global agricultural sustainability. Alongside target-site resistance (TSR), non-target-site resistance (NTSR) is becoming increasingly prevalent, complicating weed management and endangering food security. NTSR arises from physiological and biochemical modifications, including altered herbicide absorption, translocation, sequestration, and enhanced metabolic detoxification, often leading to cross-resistance across multiple herbicide modes of action. Metabolism-based resistance, primarily mediated by cytochrome P450 monooxygenases (CYP450s) and glutathione S-transferases (GSTs), has been widely reported in agriculturally significant weed species, thereby exacerbating resistance management challenges. Given the growing complexity of herbicide resistance, a comprehensive understanding of herbicide resistance mechanisms, especially NTSR, is critical for developing sustainable weed management approaches. This dissertation thesis provides current knowledge on NTSR mechanisms, highlighting recent advancements and research gaps. It outlines objectives and hypotheses focused on NTSR prevalence and mechanisms in key weed species in the Czech Republic (Apera spica-venti, Amaranthus retroflexus, and Alopecurus myosuroides). Additionally, it explores the use of omics technologies, particularly transcriptomics and metabolomics and their integration, to unravel the regulatory networks underlying NTSR. The dissertation synthesizes findings from four published studies. The results indicated that metabolism-based resistance, primarily involving CYP450s and GSTs, is the dominant NTSR mechanism in A. spica-venti and A. retroflexus. However, a co-existence of TSR and metabolism-based NTSR mechanisms was also detected in these species, highlighting the multifaceted nature of herbicide resistance evolution. In contrast, TSR, specifically target-gene mutations, was identified as the principal mechanism conferring resistance in A. myosuroides. Comparative analyses with previous studies suggest that herbicide resistance in these species is evolving towards increasingly complex, metabolically driven NTSR mechanisms. The findings presented in this dissertation enhance our understanding of the genetic and biochemical foundations of herbicide resistance, particularly through the integration of omics-based methodologies. This research contributes to the development of more effective, integrated weed management strategies tailored to the dynamic landscape of herbicide resistance in the Czech Republic and beyond. …více
