The function of Trop2 protein in cell response to stress

Abstract:

Protein Trop2 je transmembránový glykoprotein nadměrně exprimovaný u většiny epiteliálních nádorů. Jeho exprese často koreluje s agresivitou nádoru, metastazováním a špatnou prognózou. Na základě předběžných dat z naší laboratoře jsme předpokládali, že protein Trop2 může být součástí buněčné odpovědi na stres. V této práci jsme zkoumali vliv exprese proteinu Trop2 na odpověď buněk linií 4T1 a MDA-MB-231 na vybrané typy stresu (hypoxie, nízká koncentrace séra, působení chemoterapeutik). Zároveň jsme analyzovali interakci proteinu Trop2 a stresové signální dráhy JNK/AP-1. Data prezentovaná v této práci ukazují, že protein Trop2 ovlivňuje citlivost buněk k hladovění a chemoterapii, tyto efekty jsou však závislé na testované buněčné linii. Dalším zjištěním je, že exprese tohoto proteinu je negativně regulována proteiny JNK. Tato zjištění mohou vést k lepšímu pochopení regulace/funkce proteinu Trop2 v nádorových buňkách.

Abstract:

Trop2 protein is transmembrane glycoprotein which is overexpressed in most epithelial tumors. Its expression often correlates with tumor aggressiveness, metastasis and poor prognosis. Based on preliminary data obtained in our laboratory, we hypothesized that Trop2 might be involved in the cellular stress response. In this thesis, we have analyzed the effect of Trop2 expression on cell response to selected types of stress (hypoxia, low serum, chemotherapy) in 4T1 and MDA-MB-231 cells. We have also analyzed the interaction between Trop2 and JNK/AP-1 stress signalling pathway. Data presented in this thesis show that Trop2 affects cell sensitivity to starvation and chemotherapy but these effects are cell line-dependent. Furthermore, we have found that Trop2 expression is negatively regulated by JNK. These findings can lead to the better understanding of Trop2 regulation/function in cancer cells.
