Antimicrobial activity of bacteria

Abstract:

Bakalářská práce je zaměřena na mikroorganismy, konkrétně bakterie, jejich charakteristiky, struktury a tvary buněk. Dále jsou v práci popsány specializované metabolity či přírodní produkty, z hlediska jejich funkce jakožto léčiv, a strategií pro jejich další objevování. Nejvýznamnějším z nich jsou antibiotika, kterým je v práci věnována pro jejich důležitost samostatná kapitola, která zahrnuje jejich objev, klasifikaci, dělení a samotné důvody, proč jsou mikroorganismy produkovány. Navazujícím a zároveň hlavním tématem práce je antimikrobiální aktivita bakterií. Ta je hodnocena jak z pohledu kontaktní či nekontaktní formy, tak i z hlediska predace a produkce antimikrobiálních sloučenin. Uvedeny jsou i možné příklady antimikrobiální aktivity v různých prostředích. Nedílnou součástí práce je uvedení použitých laboratorních metod. Zde je zahrnuta kultivace bakterií na tekutých i pevných médiích; izolace DNA v souvislosti s využitím magnetických částic; sekvenování a metoda PCR. Cílem práce bylo identifikovat další bakterii schopnou vyrábět dosud neznámé baktericidní látky, její izolace a určení rozsahu jejího působení. Jako zdrojový materiál byl použit netopýří trus, jako zajímavý materiál dosud v podobných pracích opomíjený. Všechna tato témata jsou zahrnuta v praktické části bakalářské práce.

Abstract:

The bachelor thesis focuses on microorganisms, specifically bacteria, their characteristics, structures and cell shapes. Furthermore, the thesis describes specialized metabolites or natural products, in terms of their function as drugs, and strategies for their further discovery. The most important of these are antibiotics, which are given a chapter in the thesis due to their importance, covering their discovery, classification, division and the very reasons why they are produced by microorganisms. The antimicrobial activity of bacteria is a follow-up and the main topic of the thesis. This is evaluated both in terms of contact or non-contact form and in terms of predation and production of antimicrobial compounds. Possible examples of antimicrobial activity in different environments are also given. An integral part of the work is the presentation of the laboratory methods used. This includes the cultivation of the bacteria on liquid and solid media; DNA isolation in the context of magnetic particle use; sequencing and the PCR method. The aim of the work was to identify another bacterium capable of producing previously unknown bactericidal substances, its isolation and determination of the extent of its activity. Bat droppings were used as source material, an interesting material previously neglected in similar work. All these topics are covered in the practical part of the bachelor thesis.
