Analysis of Selected Developmental Neurotoxic Pesticides in Human and Animal Matrices

Abstract:

Celosvětově narůstá počet výskytu vývojových poruch a poruch učení u dětí. Z vědeckého hlediska není pochyb o tom, že expozice neurotoxickým chemickým látkám během raného rozvoje mozku může negativně ovlivnit učení a vývoj dětí. Projekt DENAMIC financovaný Evropskou komisí (v překladu “Hodnocení vývojové neurotoxicity směsí u dětí”) zkoumal neurotoxické účinky nízkých koncentrací směsí pesticidů a řady běžných polutantů na děti. Projekt se zaměřil na (subklinické) účinky na učení (kognitivní schopnosti) a vývojové poruchy u dětí (např. ADHD, poruchy autistického spektra a úzkostné poruchy) a bylo vynaloženo úsilí vyvinout nástroje a metody pro screening neurotoxických účinků. Vyvinuté metody a získané výsledky můžou poskytnout EU a WHO doporučení pro řízení rizik a podpořit právní předpisy EU o chemických látkách pro identifikaci (potenciálních) neurotoxických látek. Hlavním cílem této práce bylo vytvořit efektivní, rychlé a šetrné analytické metody pro stanovení neurotoxických sloučenin v malém množství biologických vzorků, jako jsou živočišné tkáně a tekutiny z in vivo studií nebo mateřské mléko a moč z kohort. Byly vyvinuty metody stanovení vybraných neurotoxických pesticidů (karbaryl, chlorpyrifos, cypermethrin, α, β-endosulfan) v tkáních potkanů a myší, jako jsou ledvinový tuk, játra, části mozku zahrnující mozkovou kůru, hipokampus, striatum, cerebellum, a v plazmě. Byla vyvinuta metoda stanovení metabolitů polybromovaných difenyletherů (PBDE) tj. hydroxylovaných PBDE a vybraných pesticidů v moči. Absolutní limit detekce pro každou sloučeninu byl na úrovni pikogramů, což nám umožnilo stanovit stopové koncentrace neurotoxických sloučenin i v malých množstvích vzorků. Výsledky analýz tkání ukázaly, že neurotoxické pesticidy (α-cypermethrin, α, β-endosulfan) jsou transportovány ze samice do mláďat a mohou působit neurotoxicky napříč generacemi. Bylo také zjištěno, že pesticidy se ukládají do tkání, i do mozku (karbaryl), ačkoli v literatuře je popsána jejich rychlá a celková eliminace z organismu. V rámci projektu DENAMIC byly získané výsledky vyhodnoceny ve spolupráci s mezinárodním týmem vědců z evropských univerzit a výzkumných institucí s cílem odhadnout rizika účinků běžně používaných pesticidů a dalších potenciálních neurotoxických sloučenin na děti, jejich prenatální vývoj a rané dětství.

Abstract:

Worldwide, serious concern has arisen about the increased incidence of learning and developmental disorders in children. From a scientific point of view, there is no doubt that exposure to neurotoxic chemicals during early brain development can adversely affect learning and development. The European commission-funded project DENAMIC "Developmental Neurotoxicity Assessment of Mixtures in Children" investigated neurotoxic effects of low-concentration mixtures of pesticides and a number of common environmental pollutants in children. The project focused on (subclinical) effects on learning (cognitive skills) and developmental disorders in children (e.g. ADHD, autism spectrum disorders and anxiety disorders) and developed tools and methods for screening of neurotoxic effects. The developed methods and obtained results could give recommendations for risk management to the EU and WHO and support the EU chemicals legislation for identifying (potential) neurotoxicants. Main goal of our work was to create effective, quick and cheap analytical tools for determination of neurotoxic compounds in different types of limited biological samples, such as animal tissues and fluids from in vivo studies or breast milk and children’s urine from mother-child cohorts. The analytical procedures for determination of group of neurotoxic pesticides (carbaryl, chlorpyrifos, cypermethrin, α- and β-endosulfan) in small amount of mice and rat tissues, such as perirenal fat, liver, parts of brain - cerebral cortex, hippocampus, striatum, cerebellum, and plasma have been developed. The method for determination of metabolites of polybrominated diphenyl ethers (PBDEs) i.e. hydroxylated PBDEs, and selected pesticides in urine was developed. The absolute limits of detection for each compound were in pg levels, which allowed us to measure trace concentrations of pollutants in amounts of samples. The results of analyses shown that neurotoxic pesticides (α-cypermethrin, α- and β-endosulfan) are transported from rat mothers to their pups, and thus can act neurotoxically through generation. Moreover, pesticides are cumulated in tissues, e.g. into the brain (carbaryl), although their rapid and total elimination from the organism is described in the literature. Within the DENAMIC project, the obtained results were evaluated in cooperation with international scientific teams from European universities and research institutions in order to estimate risks of effects of commonly used pesticides and other potential neurotoxic compounds on children who are exposed them during prenatal development and early childhood.
