\kur{Plectranthus amboinicus} and its antimicrobial effects

Abstract:

Tato diplomová práce pojednává o antimikrobiálních účincích byliny Plectranthus amboinicus. Hlavní kapitoly jsou věnovány taxonomii, morfologii, chemickému složení, účinkům a využití rodu Plectranthus a převážně druhu Plectranthus amboinicus, u kterého jsou popsány těkavé a netěkavé chemické složky a zároveň také antibakteriální a antimykotické účinky extraktů a esenciálního oleje. V experimentální části byl z rýmovníku vytvořen nejen esenciální olej, ale i extrakty z čerstvých a lyofilizovaných listů pomocí ultrazvuku v různých extrakčních rozpouštědlech (96% ethanol, 60% ethanol a horká destilovaná voda). Následně byla u vybraných grampozitivních a gramnegativních bakterií a jednoho druhu kvasinky testována antimikrobiální účinnost extraktů a esenciálního oleje z rýmovníku pomocí difuzního agarového testu. Další část pojednává o testování minimální inhibiční koncentrace pomocí bujónové mikrodiluční volatilizační metody a také o analýze chemického složení esenciálního oleje z byliny Plectranthus amboinicus.

Abstract:

This diploma thesis discusses the antimicrobial effects of Plectranthus amboinicus. The main chapters are devoted to taxonomy, morphology, chemical composition, effects and use of genus Plectranthus and mainly Plectranthus amboinicus, in which volatile and non-volatile chemical components are described and also antibacterial and antifungal effects of extracts and essential oil. In the experimental part, not only essential oil but also extracts from fresh and lyophilized leaves were created from the rhyme tree using ultrasound in various extraction solvents (96% ethanol, 60% ethanol and hot distilled water). Subsequently, the antimicrobial efficacy of extracts and essential oil from the rhyme tree was tested using a diffuse agar test in selected gram-positive and gram-negative bacteria and yeast. The next part deals with the testing of the minimum inhibitory concentration using the broth microdilution volatilization method and also the analysis of the chemical composition of the essential oil from the herb Plectranthus amboinicus.
