Effect of bovine serum albumin on the degradation of hyaluronan using hyaluronidases

Abstract:

Bakalářská práce se zaměřuje na modulaci štěpení kyseliny hyaluronové pomocí přídavku bovinního sérového albuminu, který je schopen interagovat s kyselinou hyaluronovou a ovlivňovat průběh její degradace hyaluronidázami. Sledování rychlosti enzymatické katalýzy kyseliny hyaluronové po přídavku bovinního sérového albuminu bylo studováno pomocí kolorimetrické Morgan-Elsonovy metody. Porozumění tvorby elektrostatických komplexů mezi kyselinou hyaluronovou a bovinním sérovým albuminem bylo studováno jak na základě tvorby zákalu pomocí turbidimetrie, tak na základě jejich vazebné afinity pomocí molekulárního dokování. Z provedených experimentů vyplývá, že bovinní sérový albumin je schopen zvyšovat počáteční rychlost enzymové degradace kyseliny hyaluronové v prostředí s nízkou iontovou silou. Pomocí turbidimetrických experimentů byla potvrzena tvorba komplexů kyseliny hyaluronové a bovinního sérového albuminu. Molekulární dokování predikovalo větší vazebnou afinitu kyseliny hyaluronové k bovinnímu sérovému albuminu než k hyaluronidáze. Nicméně na základě nízkého rozdílu vazebných afinit nelze data jednoznačně interpretovat.

Abstract:

The bachelor thesis focuses on the modulation of the cleavage of hyaluronic acids by the addition of a bovine serum albumin, which is able to interact with hyaluronic acid and influence the course of its degradation by hyaluronidases. Monitoring the rate of enzymatic catalysis of hyaluronic acid after the addition of bovine serum album was studied using the Morgan-Elson colorimetric method. The understanding of the formation of electrostatic complexes between hyaluronic acid and bovine serum album was studied both on the basis of turbidimetry formation by turbidimetry and on the basis of their binding affinity by molecular docking. Experiments show that bovine serum albumin is able to increase the initial rate of enzymatic degradation of hyaluronic acid at low ionic strength environment. The formation of complexes of hyaluronic acid and bovine serum album was confirmed by turbidimetric experiments. Molecular docking predicted a higher binding affinity of hyaluronic acid for bovine serum albumin than for hyaluronidase. However, due to the low difference in binding affinities, the data cannot be interpreted unambiguously.
