Antimicrobial activity of honey bee haemolymph

Abstract:

Hlavní obranná reakce hmyzu proti patogenům je realizovaná prostřednictvím antimikrobiálních faktorů. Úvodní část práce obecně seznamuje s působením antimikrobiálních faktorů probíhající v hemolymfě hmyzu. Důraz je však kladen na podrobnější popis a působení antimikrobiálních faktorů v hemolymfě včel Apis mellifera. Ke studiu antimikrobiální aktivity byla použita hemolymfa získaná ze dvou experimentálních organismů. Hemolymfa získaná z larev Galleria mellonella VII. instaru sloužila pro optimalizaci měření. Parametry optimalizačního měření byly použity pro měření hemolymfy získané z dělnic včely medonosné. Antimikrobiální aktivita hemolymfy byla primárně stanovována za využití luminometrické metody. Antimikrobiální účinek hemolymfy byl testován na bioluminiscenčních bakteriích Escherichia coli K12. Měření antimikrobiální aktivity hemolymfy bylo rozšířeno o kultivační a spektrofotometrické stanovení. Luminometrickou metodou bylo prokázáno, že antimikrobiální aktivita hemolymfy včely medonosné je nižší v porovnání s hemolymfou Galleria mellonella.

Abstract:

The main defense response against pathogens of insects is realized through antimicrobial factors. The first part of this thesis introduces the mechanisms of action of antimicrobial factors occurring in insect haemolymph. The emphasis is laid on the description and effects of antimicrobial factors in the haemolymph bee Apis mellifera. The antimicrobial activity was studied in insect haemolymph which was obtained from two experimental organisms. The haemolymph obtained from Galleria mellonella larvae of VII. instar was used to optimize the antimicrobial assays. The parameters from the optimization measurements were used to measure the antimicrobial activity in hemolymph of the worker honeybees. The antimicrobial activity of the haemolymph was determined mostly by luminomteric method. The antimicrobial effect of the haemolymph was tested on the viability of Escherichia coli K12 which has the ability to emit light. Cultivation and spectrophotometric methods were used to compare the results. Luminometric method has been shown that the antimicrobial activity of the bee haemolymph is less effective compared to the haemolymph of Galleria mellonella.
