Synthesis of gold nanoparticles with emphasis on their shape and size for biomedical applications

Abstract:

Využití nanomateriálů se rozrůstá i do prostředí medicínské diagnostiky a terapie, kde nachází potenciál pro zvýšení její efektivity a snížení nežádoucích účinků použitých léčiv. Důležitým příkladem je cílený transport cytostatik v rámci chemoterapie při léčbě rakoviny. Tato práce se zabývá biomedicínskými aplikacemi nanomateriálů, především aplikacemi nanočástic zlata, které jsou významnými kandidátními materiály pro cílený transport léčiv, hojení ran, zvýšení antibakteriálního účinku antibiotik, lékařskou diagnostiku a teranostiku. V praktické části této práce je detailně popsané provedení syntézy zlatých nanočástic různých tvarů a velikostí s povrchovou modifikací. Zejména tyčinek, sférických částic a dalších tvarů, jejich výsledná charakterizace a vizualizace. V praktické části této práce byly připraveny nanočástice na bázi zlata (AuNP) různých tvarů a velikostí s povrchovou modifikací cetyltrimethylammonium-bromidem (CTAB). Nanočástice byly připraveny pomocí metody dvoufázové syntézy, byly charakterizovány metodou dynamického rozptylu světla (DLS) a měřením zeta potenciálu na přístroji ZetaSizer Nano ZS (Malvern Instruments Ltd, VB). Vizualizace AuNP byla vykonána pomocí skenovacího elektronového mikroskopu (SEM). Podařilo se připravit, charakterizovat a vizualizovat sférické AuNP, AuNP ve tvaru tyčinek a bipyramid a také dvě směsi, které obsahovaly jak nanočástice, tak submikronové částice zlata různých tvarů a velikostí. AuNP ve tvaru tyčinek, bipyramidy i sférické částice nachází možné uplatnění v biomedicínských aplikacích, v oblasti fotokatalýzy, spektroskopie aj.

Abstract:

The use of nanomaterials has been recently expanding into the area of medical diagnostics and therapy, where a potential to increase its efficiency and reduce the side effects of the drugs used can be found. An important example is the targeted transport of cytostatics in chemotherapy by cancer treatment. This thesis is focused on biomedical applications of nanomaterials, especially applications of gold nanoparticles, which are important candidate materials for targeted drug transport, wound healing, enhancement of the antibacterial effect of antibiotics, medical diagnostics and theranostics. In the practical part of this thesis, the synthesis of gold nanoparticles of different shapes and sizes with surface modification is described in detail. In particular, bars, spherical particles and other shapes, their resulting characterization and visualization. In the practical part of this work, gold nanoparticles (AuNPs) of various shapes and sizes with surface modification with cetyltrimethylammonium bromide (CTAB) were prepared. Nanoparticles were prepared using a two-phase synthesis method, characterized by dynamic light scattering (DLS) and zeta-potential measurements on a ZetaSizer Nano ZS (Malvern Instruments Ltd, UK). AuNPs were visualized using a scanning electron microscope (SEM). Spherical AuNPs, rod-shaped AuNPs, and bipyramids, as well as two mixtures containing both nanoparticles and submicron gold particles of various shapes and sizes, were successfully prepared, characterized, and visualized. AuNPs in the form of rods, bipyramids, and spherical particles find possible applications in biomedical applications, in the field of photocatalysis, spectroscopy, etc.
