Polymerase chain reaction as a tool for detection and differentiation of the causative agent of American foulbrood (Paenibacillus larvae)

Abstract:

Mor včelího plodu, jehož původcem je bakterie Paenibacillus larvae, je závažné onemocnění larev včely medonosné. Choroba má negativní dopad na včelaře a zejména pěstování řady úrodných plodin. Rychlá detekce patogenu a diferenciace genotypů je důležitá pro správnou volbu eliminace. Tato práce se zabývá vývojem detekčního a diferenciačního systému založeného na metodě real time PCR umožňující vysoce specifickou analýzu v krátkém čase. Z testovaných návrhů bylo nejlepšího výsledku dosaženo detekčním systém PENNY. Při cut-off hodnotě Cq = 35 systém prokázal senzitivitu 90 % a také dobrou specifitu. Ta je dána negativním výsledkem Paenibacillus alvei.

Abstract:

American foulbrood, caused by bacterium Paenibacillus larvae, is a serious infectious disease of honey bee larvae with a negative impact on beekeepers , and particularly, on crops cultivation. Rapid pathogen detection and genotype differentiation is essential for effective elimination. Thus, this thesis deals with the development of detection and differentiation assay based on the real time PCR method, which is highly specific and allows pathogen identification in a short time. The best results were received using the PCR detection system PENNY. With a cut-off value of Cq = 35, this system achieved sensitivity of 90 %. The high specifity of the system was demonstrated by the negative result of bacterium Paenibacillus alvei.
