Effect of emetine on DNA replication in human cancer cell lines

Abstract:

Předkládaná bakalářská práce se zabývá vlivem emetinu na replikaci DNA u lidských nádorových buněk. Emetin je v současné době považován za specifický inhibitor syntézy opožďujícího se vlákna DNA. Literární část práce obsahuje popis replikace DNA se zaměřením na syntézu opožďujícího se vlákna, souhrn působení emetinu a také se věnuje popisu rozpojení syntézy vedoucího a opožďujícího se vlákna DNA. Cílem experimentální části této práce bylo ověření schopnosti emetinu zablokovat syntézu opožďujícího se vlákna DNA. K analýze proliferace buněk po ovlivnění emetinem byla využita CLICK reakce, která spočívá v detekci inkorporace analogu thyminu 5-ethynyl-2´-deoxyuridinu do replikující se DNA. Bylo zkoumáno různé časové a koncentrační ovlivnění buněk emetinem. Ze získaných výsledků vyplývá výrazný pokles míry replikace již po krátkém působení emetinu. Dále byla stanovována hladina jednovláknových řetězců DNA, k jejichž akumulaci by při zablokování syntézy opožďujícího se vlákna DNA mělo v buňkách docházet. K tomuto účelu byly využity dvě odlišné metody, které jsou založené na nepřímém imunofluorescenčním barvení buněk. Jedna metoda vychází z analýzy inkorporace analogu thyminu 5-bromo-2´-deoxyuridinu do jednovláknových řetězců DNA, druhá metoda je založená na detekci replikačního proteinu A navázaného na jednovláknové řetězce DNA. Pro porovnání účinků emetinu na syntézu opožďujícího se vlákna byl v těchto experimentech použit inhibitor Pol , známý pod označením ST1926. Výsledky získané použitím těchto dvou látek se signifikantně liší, z čehož byl vyvozen závěr, že emetin nepůsobí jako specifický inhibitor syntézy Okazakiho fragmentů.

Abstract:

This bachelor thesis deals with effect of emetine on DNA replication in human cancer cell lines. Emetine is currently considered as specific inhibitor of lagging strand synthesis. The literature part of this thesis contains description of DNA replication with a focus on lagging strand synthesis, a summary of the biological effects of emetine and also describes an uncoupling of leading and lagging strand synthesis. The aim of the experimental part of this thesis was to verify the ability of emetine to inhibit the lagging strand synthesis. The CLICK reaction, which is based on detection of thymine analogue 5-ethynyl-2´-deoxyuridine into replicating DNA, was used to analyse cell proliferation after treatment with emetine. Various time exposition and concentration effects of emetine on cell proliferation were measured. The obtained results show a significant decrease in replication rate even after a short exposure to emetine. In the next part of the thesis the level of single-stranded DNA was measured, because their accumulation in cells should occur when the lagging strand synthesis is blocked. For this purpose two different methods were used, both are based on indirect immunofluorescence staining of cells. One method is based on detection of incorporated thymine analogue 5-bromo-2´-deoxyuridine into a single stranded DNA under non-denaturing conditions. The second one is based on the detection of replication protein A bounded to a single stranded DNA. To compare the effects of emetine on lagging strand synthesis the Pol inhibitor, known as ST1926, was used. The results obtained using these two substances differ significantly, leading to conclusion that emetine is not a specific inhibitor of Okazaki fragment synthesis.
