Molecular capsules for metallodrugs: Towards novel antitumor therapies

Abstract:

Vývoj účinných nosičů léčiv je důležitou oblastí farmaceutického výzkumu. Tato diplomová práce se zabývá přípravou nosičů odvozených od biokompatibilních cyklodextrinů se zaměřením na vývoj molekulárních kapslí, inkluzních komplexů a nanočástic metalo-organických sítí. Našim cílem je aplikovat tyto nosiče na enkapsulaci a transport metaloléčiv s možnými protirakovinnými účinky, což může usnadnit cílení léčiva do rakovinných tkání, poskytnout možnost selektivního uvolnění léčiva v kyselejším a vysoce redukčním prostředí rakovinných buněk a výrazně zlepšit rozpustnost metaloléčiv ve vodném prostředí, a tedy i ve fyziologickém prostředí lidského těla. V rámci experimentální části diplomové práce jsme se nejprve zabývali tvorbou inkluzních komplexů, kde se nám potvrdila dobrá afinita metaloléčiv k cyklodextrinům. Poté jsme se zabývali přípravou dimerních molekulárních schránek, přičemž jsme vycházeli z různých derivátů β- a γ-cyklodextrinu. A nakonec jsme se věnovali syntéze porézních metalo-organických sítí, které mají schopnost adsorbovat metaloléčivo do svých struktur.

Abstract:

The development of effective drug carriers is an important area of pharmaceutical research. This thesis deals with the preparation of carriers derived from biocompatible cyclodextrins focus on the development of molecular capsules, inclusion complexes and nanoparticles of metal-organic networks. Our goal is to apply these carriers to the encapsulation and transport of metallo-drugs with possible anti-cancer effects, which can facilitate drug targeting into cancer tissues, provide the possibility of selective drug release in the more acidic and highly reducing environment of cancer cells, and significantly improve the solubility of metallo-drugs in aqueous media and hence in the physiological environment of the human body. In the experimental part of the thesis we first dealt with the formation of inclusion complexes, where we confirmed the good affinity of metallo-drugs for cyclodextrins. Then we dealt with the preparation of dimeric molecular boxes, starting from various derivatives of β- and γ-cyclodextrin. Finally, we have devoted ourselves to the synthesis of porous metal-organic networks, which have ability to adsorb metal-drug into their structures.
