Chromatin plasticity and epigenetics in embryonic stem cells

Abstract:

This thesis deals with the chromatin plasticity and epigenetics in embryonic stem cells. For better understanding, the morphology and bacis characteristics of embryonic stem cells are mentioned at the beginning. Self renewal and pluripotency are chosen as the key properties, and that is why in the next chapters, the factors of pluripotency are divided into three groups and each of them is described in more details. However, the effect of those factors on genes is influenced by the level of chromatin compactness, which is regulated by epigenetic changes. The main part of thesis describes epigenetic changes in histones, which are somehow connected to the other epigenetic factors, like DNA methylation. Closing part of thesis is dedicated to discussion on the correlation between chromatin plasticity and stem cell potency, with the emphasis on HP1 protein.

Abstract:

V této bakalářské práci je rozebraná plasticita chromatinu a epigenetika embryonálních kmenových buněk. Pro lepší pochopení je v první části popsaná základní charakteristika kmenových buněk a jejich morfologie. Jako klíčové vlastnosti jsou určené schopnost sebeobnovy a pluripotence. Z tohoto důvodu jsou faktory pluripotence rozdělené do třech skupin, přičemž každá z nich je detailně popsaná. Účinek těchto faktorů na samotné geny je ovlivněný úrovní sbalení chromatinu, která je regulovaná epigenetickými změnami. V práci je kladen velký důraz na epigenetické úpravy histónů, které jsou provázány s ostatními epigenetickými znaky, jako například metylace DNA. Nakonec je v závěru práce diskutovaná korelace mezi plasticitou chromatinu a potenčním stavem kmenových buněk, s vyzdvihnutím důležitosti HP1 proteinu.
