Biomarker responses to toxic cyanobacteria in aquatic vertebrates and birds

Abstract:

Rigorózní práce se zabývá negativním působením sinic (cyanobakterií) a jejich toxinů na antioxidační a detoxikační systém vodních obratlovců a ptáků. Práce se skládá ze dvou propojených částí: přehled biomarkerů toxického působení sinic na vodní obratlovce a experimentální studie s modelovým organismem křepelkou japonskou. První část shrnuje současné poznatky o biomarkerech negativního působení sinic ( komplexní biomasy a čistých cyanotoxinů – microcystinů) u sladkovodních obratlovců se zaměřením na biomarkery detoxikace a oxidativního stresu. Mimo to se podrobně zaměřuje na obecné trendy změn těchto parametrů a faktory limitující jejich praktické využití. Druhou část tvoří experimentální studie negativních efektů působení sinic na ptáky v koexpozicích s dalšími stresory prostředí (olovo a patogenní mikroorganismy ve formě vakcíny). Konkrétně se jedná o studii změn parametrů detoxikace a oxidativního stresu u modelového organismu – křepelky japonské. Uspořádání testu s více stresory lépe modeluje reálné situace v prostředí a přináší tak unikátní informace o efektech tohoto spolupůsobení na sledované parametry. Výsledky této studie ukázaly významné ovlivnění studovaných parametrů a to především u ptáků vystavených působení koexpozic, což potvrzuje hypotézu o možné kombinaci negativních účinků a zvýšení celkové toxicity působících stresorů.

Abstract:

This rigorous thesis is addressing negative effects of cyanobacteria and their toxins on antioxidative and detoxification system of aquatic vertebrates and birds. It consists of two connected parts: a review of biomarker responses to toxic cyanobacteria in aquatic vertebrates and an experimental study with model bird Japanese quail. First part summarizes existing information about biomarker responses observed in freshwater aquatic vertebrates after exposures to both isolated cyanobacterial toxins - microcystins and complex cyanobacterial sample with special attention to biomarkers of detoxification and oxidative stress. Several general trends and factors that limit practical applications of these parameter responses are discussed in detail. Second part is an experimental study focused on negative effects of cyanobacteria in co-exposure with others stressors (lead and immunological challenge) on the parameters of detoxification and oxidative stress in model birds – Japanese quails. The test design with multiple stressors reflects the real environmental situation and brings unique information on the effects of combined avian exposure on antioxidation parameters. The results documented significant modulations of sublethal parameters in the exposed birds mostly in the combined exposure groups and confirmed the hypothesis that adverse effects of toxic cyanobacteria with other stressors may combine and lead to higher toxicity.
