Heavy Metal Nephrotoxicity

Abstract:

V úvodu práce jsou stručně charakterizovány těžké kovy a jejich vliv na životní prostředí. Hlavní část textu je zaměřena na kadmium, rtuť a olovo, u kterých jsou popsány způsoby expozice, mechanismy transportů do buněk a toxické účinky na organismy. Těžké kovy patří mezi silně nefrotoxické látky ovlivňující funkci a stavbu ledvin. Nefrotoxické působení těžkých kovů lze sledovat pomocí in vivo či in vitro studií. V práci jsou stručně zmíněny in vitro způsoby testování nefrotoxicity se zaměřením na vhodné buněčné linie. Nejpoužívanější buněčnou linií jsou HK-2 buňky, pocházející z ledvin zdravého dospělého člověka. Často se používají při zjišťování mechanismu nefrotoxického působení cizorodých látek na funkci ledvin, mechanismu transportu cizorodých látek do ledvinných buněk a při identifikaci transportérů zapojujících se do přenosu cizorodých látek v ledvinách. Mezi další lidské buněčné linie často používané při výzkumu nefrotoxicity patří například RPTEC/TERT1 buněčná linie. Dále jsou ve výzkumu hojně využívány i zvířecí buněčné linie NRK-52E, JTC-12, WKPT, OK, LLC-PK1 a MDCK, které jsou v práci také zmíněny.

Abstract:

I briefly summarized heavy metals and their influence on the environment in this thesis introduction. The main part of this thesis is focused on cadmium, lead and mercury. By these elements are described ways of exposition, transport mechanisms to cells and toxic effects to organism. Heavy metals are highly nephrotoxic substances that affect kidney function and structure. The nephrotoxic effect of heavy metals can be monitored by in vivo or in vitro studies. In vitro methods in this thesis are mentioned in the context of nephrotoxicity testing focus on a suitable cell line. The most widely used cell line is HK-2 cell line. HK-2 cells come from kidneys of a healthy adult person. They are often used to detect of the mechanism of nephrotoxic effect of strange substances on kidney function, the transport mechanism of strange substances in kidney cells, and to identification of transporters involved in the transfer of strange substances in kidneys. Other human cell linesused mainly in nephrotoxicity research include for example, RPTEC/TERT1 cell lines. Furthermore, the animal cell lines NRK-52E, JTC-12, WKPT, OK, LLC-PK1 and MDCK, which are also mentioned in this thesis, are widely used.
