Modern methods for detection of patogens in water.

Abstract:

Bakalářská práce {\clqq}Moderní metody detekce patogenů v pitné vodě`` je zaměřená na problematiku mikrobiální kontaminace pitné vody. Úvodní kapitola se věnuje obecnému popisu, rozdělení a využití vod. První část práce je shrnutím nejvýznamnějších mikroorganismů, vyskytujících se ve vodách. Mikroorganismy jsou zde rozděleny podle hygienického a zdravotního významu na indikátory obecného znečištění, fekálního znečištění a na mikroorganismy hygienicky významné. Pro každou skupinu mikroorganismů jsou uvedeny klasické metody stanovení i moderní metody, které popisuje odborná literatura. Druhá část práce začíná úvodem do genetiky {--} vysvětlením základních termínů, a pokračuje popisem metody PCR (polymerázová řetězová reakce) a jejích modifikací. Konkrétněji jsem se zaměřila na metodu real-time PCR, její průběh, výhody i nevýhody. Tato metoda patří k moderním velmi přesným a celosvětově rozšířeným metodám jejichž využití v rámci detekce patogenů v pitné vodě je na vzestupu. Závěr shrnuje možnosti této poměrně nové molekulárně biologické metody, kterou lze použít v mnoha oborech {--} např. ve zdravotnictví (určení genetických vad a onemocnění, detekce patogenů v klinických materiálech), k průkazu mikroorganismů ve vzorcích vody či v kriminalistice (fingerprinting).

Abstract:

Thesis {\crqq}Modern methods of detection of pathogens in drinking water`` is focused on microbial contamination of drinking water. Introductory chapter deals with the general description, distribution and use of water. The first part is a summary of the most important microorganisms occurring in the waters. Microorganisms are classified according to hygienic and medical importance of the indicators of general pollution and faecal contamination of hygienically relevant microorganisms. For each group of microorganisms there are presented classical and modern methods of measurement methods, as described in the expert literature. The second part begins with an introduction to genetics - an explanation of basic terms, and goes on to describe PCR - polymerase chain reaction and its modifications. More specifically, I focused on the method of real-time PCR, its course, advantages and disadvantages. This method belongs to very accurate and widespread used methods whose detection of pathogens in drinking water is on the rise. The conclusion summarizes these relatively new molecular biological methods, which can be used in many fields - such as health care (identification of genetic defects and disease, detection of pathogens in clinical materials) for the detection of microorganisms in water samples and in criminology (fingerprinting).
