Identification of powdery mildew resistance genes in Hordeum vulgare using DNA markers

Abstract:

Padlí travní způsobené houbovým patogenem Blumeria graminis f. sp. hordei (Bgh) je jednou z nejvýznamnějších a nejrozšířenějších chorob ječmene, která snižuje celkový výnos i kvalitu zrna ječmene a tím i rentabilitu pěstování této plodiny. Z ekonomického i ekologického hlediska je pro pěstitele nejvýhodnější a nejefektivnější ochranou před patogenem pěstování odolných odrůd. Planý ječmen (Hordeum vulgare ssp. spontaneum), v jehož kolekci bylo zjištěno několik nových plně účinných zdrojů odolnosti k evropským patotypům padlí travního, je vhodným donorem genů rezistence využitelných ve šlechtění ječmene. Zdroj PI466197 byl studován s cílem určit počet genů determinujících odolnost, jejich typ dědičnosti a vztah k lokusu Mla a lokalizovat je do genetické mapy ječmene pomocí DNA markerů. Genetické i molekulární analýzy byly prováděny s rostlinami populace F2 vzniklé křížením odrůdy Tiffany nesoucí neúčinné geny odolnosti Mla7 a MlaMu2 se zdrojem odolnosti PI466197 získaného z genové banky USDA. Testy rezistence byly provedeny na Zemědělském výzkumném ústavu Kroměříž, s.r.o. a výsledky genetické analýzy se 462 rostlinami F2 ukázaly, že rezistence pocházející z tohoto zdroje je geneticky podmíněna dvěma geny s dominantním typem dědičnosti a testem na alelizmus bylo potvrzeno, že jeden gen se nachází v lokusu Mla. Předmětem molekulární analýzy bylo určit pozice genů rezistence na chromozómech ječmene a najít DNA markery v co nejtěsnější vazbě. Pro detekci genů a genetické mapování byly využity markery SSR (Simple Sequence Repeat) vybrané z literárních zdrojů a on-line databází. Pro každý marker byl optimalizován amplifikační cyklus a ověřen polymorfizmus mezi rodiči. Pro oblasti genomu s nedostatkem polymorfních mikrosatelitních markerů byly dále vybrány markery STS (Sequence-Tagged Site) a markery odvozené ze sekvence genu RGH1a v lokusu Mla, které byly konvertovány na markery typu CAPS (Cleaved Amplified Polymorphic Sequence). Primery pro tyto markery byly převzaty z databáze GrainGenes nebo navrženy v programu Primer3 ze sekvence daného markeru STS. Po optimalizaci amplifikačního cyklu byl testován polymorfizmus mezi rodiči po štěpení produktů vhodnými restrikčními enzymy. Pomocí fenotypových krajních tříd rezistentních a náchylných rostlin generace F2 a mikrosatelitních markerů rovnoměrně rozmístěných na všech chromozómech ječmene byla zachycena vazba s markery na chromozómech 1H a 2H. Statistická významnost vazby byla ověřena pomocí dat získaných vyhodnocením 113 rostlin všech genotypů v programu Map Manager QTX, který také určil pořadí a vzdálenosti mezi geneticky vázanými markery. Jeden gen rezistence byl lokalizován na krátkém raménku chromozómu 1H (lokus Mla) mezi markery RGH1aE1 a Bmac0213, 4 cM od markeru RGH1aE1 a 10 cM od Bmac0213. Pozice druhého genu na krátkém raménku chromozómu 2H byla určena mezi markery Bmac0134 a MWG878 ve vzdálenosti 4 a 8 cM. V této oblasti zatím nebyl identifikován žádný gen odolnosti pocházející z Hordeum vulgare ssp. spontaneum. Tento gen proto představuje potenciální nový gen rezistence s velkým účinkem využitelný v kombinaci s genem Mla nebo jinými geny rezistence ve šlechtění ječmene.

Abstract:

Powdery mildew caused by the fungus Blumeria graminis f. sp. hordei (Bgh) is one of the most common and wide-spread foliar diseases of barley in temperate climates including the Czech Republic. Powdery mildew can have a considerable effect on barley production because it reduces its grain yield and quality. An effective and economical way to protect the crop against the pathogen is to breed and grow resistant varieties. There is only a limited number of resistance genes in cultivated barley, but wild barley (Hordeum vulgare ssp. spontaneum) represents a promising source of new resistance genes. The accession PI466197 of wild barley was studied with the aim to characterize the genes conferring resistance to powdery mildew and localize them on a barley genetic map using DNA markers. The genetic as well as the molecular analyses were performed in the segregating F2 population of the cross between the winter barley cultivar Tiffany possessing the ineffective genes Mla7 and MlaMu2 and the resistant accession PI466197 from the USDA National Small Grains Collection. The resistance tests were carried out at the Agricultural Research Institute in Kroměříž and the genetic analysis with 462 F2 plants showed that the resistance in this accession was determined by two independent genes with dominant mode of inheritance, one of the genes being in the Mla locus. The object of the molecular analysis was to localize the identified resistance genes on barley chromosomes and find tightly linked DNA markers. Microsatellite (SSR – Simple Sequence Repeat) markers, CAPS (Cleaved Amplified Polymorphic Sequences) markers derived from barley STS (Sequence-Tagged Site) from on-line databases and literature, and markers designed from the RGH1a gene sequence of the Mla locus were used for genetic linkage detection and mapping. Primers for the markers were taken from the GrainGenes database or generated in the Primer3 program using the known STS marker sequence. The amplification cycles for the markers were optimized and polymorphism between the parents was investigated based on the differences in the length of microsatellites or DNA fragments cleaved by restriction enzymes. Using microsatellite markers and bulks of resistant and susceptible F2 plants, the resistance genes were localized on chromosomes 1H and 2H. Data from 113 F2 plants of all genotypes were analysed in the Map Manager QTX program to confirm the linkage statistically and determine the order and distances between the markers. One resistance gene was localized on the short arm of chromosome 1H between the markers RGH1aE1 and Bmac0213, 4 cM from RGH1aE1 and 10 cM from Bmac0213. The position of the resistance gene on the short arm of chromosome 2H was established between the markers Bmac0134 and MWG878, in the distance of 4 and 8 cM, respectively. Until now, no gene conferring powdery mildew resistance originating from H. vulgare has been located on the short arm of barley chromosome 2H, so it could be a newly identified locus of powdery mildew resistance. These results can offer new possibilities in barley breeding. The markers tightly linked to the resistance genes can be used for marker-assisted selection and gene pyramiding.
