Abstract:

Výzkum metaloléčiv na bázi platiny se neustále rozvíjí s cílem překonat omezení tradičních látek podobných cisplatině a zvýšit jejich terapeutickou účinnost. V této práci bude studována interakce hostitel-host mezi proléčivy na bázi platiny(IV) odvozenými od oxaliplatiny, monofunkčními Pt(II) deriváty cisplatiny a makrocyklickými nosiči, jako jsou cyklodextriny a kukurbiturily. K vyhodnocení vazebných afinit těchto komplexů byly použity 1H NMR titrační experimenty. Způsob interakce komplexů platiny(II) s kanonickými duplexními a G-kvadruplexními strukturami DNA byl charakterizován pomocí 1H NMR titrace a 1H-1H 2D NOESY experimentů. Výsledkem je zjištění, že monovalentní komplex platiny(IV) vykazuje vyšší afinitu ke kukurbit[7]urilu než β-cyklodextrinu. Jeho derivát, divalentní komplex, při tvorbě dienkapsulovaných komplexů vykazoval nekooperativní vazbu β-cyklodextrinu na monoenkapsulovaný komplex a homotropní negativní kooperativitu vazby dvou kukurbit[7]urilů. Zapouzdření komplexů platiny(II) s kukurbit[7]urilem bylo charakterizováno středně silnou vazebnou afinitou, přičemž u komplexu PtPyrPhe došlo v přítomnosti kukurbit[7]urilu k rychlé hydrolýze. Studium PtPyrPhe se sekvencemi DNA prokázalo specifickou vazbu do žlábku v AT oblastech duplexu DNA a slabou interakci nad horní G-tetrádou G-kvadruplexu.

Abstract:

The research of platinum-based metallodrugs is constantly developing to overcome the limitations of traditional cisplatin-like agents, enhancing their therapeutic efficacy. In this work, the host-guest interaction of oxaliplatin-derived platinum(IV) prodrugs and cisplatin-derived monofunctional platinum(II) complexes was studied with macrocyclic carriers such as cyclodextrins and cucurbiturils. The 1H NMR titration experiments were used to evaluate the binding affinities of these complexes. The interaction mode of platinum(II) complexes with canonical duplex and G-quadruplex DNA structures was characterised using 1H NMR titration and 1H-1H 2D NOESY experiments. As a result, the monovalent platinum(IV) complex exhibited a higher affinity for cucurbit[7]uril over β-cyclodextrin. Its derivative, divalent complex, in the formation of diencapsulated complexes, revealed the non-cooperative binding of β-cyclodextrin to the monoencapsulated complex and homotropic negative cooperativity of the binding of two cucurbit[7]urils. The encapsulation of platinum(II) complexes with cucurbit[7]uril showed moderate affinity bindings, with the phenylpyridine-based Pt complex (PtPyrPhe) complex experiencing rapid hydrolysis in the presence of cucurbit[7]uril. The study of PtPyrPhe with DNA sequences demonstrated specific groove binding at the AT regions of the DNA duplex and weak interaction on top of the top G-tetrad of the G-quadruplex.
