Působí pozitivní selekce na geny specificky exprimované v samčích rostlinách rodu Silene ?

Abstract:

U člověka a šimpanze bylo pro prokázanou adaptivní evoluci genů exprimovaných v samčích pohlavních orgánech navrženo vysvětlení na základě vnitrogenomového konfliktu vyvolaného kompeticí zárodečných buněk během spermatogeneze. Cílem této diplomové práce bylo zjistit, zda se analogické geny Silene latifolia a Silene diclinis, u nichž ke kompetici během mikrosporogeneze nedochází, od divergence těchto dvou druhů rovněž vyvíjely pod vlivem pozitivní selekce. U dvou pětin celkového počtu analyzovaných ETS ze samčích pohlavních orgánů S. latifolia byly nalezeny homology u Arabidopsis thaliana, které však nevykazovaly expresi specifickou pro prašníky. Většina analyzovaných ETS nemá u A. thaliana žádné homology. Jejich pohlavně specifická funkce je tedy zřejmě evolučně poměrně mladá. Expresní analýzou pomocí RT-PCR bylo zjištěno, že jen 6 ze 40 EST, pro které byla publikována exprese specifická pro prašníky, je skutečně exprimováno specificky v prašnících. Exprese 20 EST vykazovala odlišnosti závislé na pohlaví. Srovnáním sekvencí 6 genů, jejichž exprese byla specifická pro prašníky nebo samčí rostliny, a 9 genů, jejichž expresní profily naznačovaly možnou souvislost se samčí fertilitou, mezi druhy S. latifolia a S. diclinis nebyly zjištěny známky probíhající adaptivní evoluce, sekvence se naopak zdají být výrazně konzervovanější než sekvence genů bez vztahu k samčím pohlavním orgánům. Geny exprimované v samčích pohlavních orgánech dvoudomých rostlin rodu Silene pravděpodobně adaptivní evolucí prošly před divergencí druhů S. latifolia a S. diclinis, ale od jejich divergence se měnily jen velmi málo nebo vůbec a pozitivní selekce na ně v současné době nepůsobí. Vnitrogenomového konflikt vyvolaný kompeticí zárodečných buněk během spermatogeneze tedy zůstává uspokojivým vysvětlením pozitivní selekce genů exprimovaných v samčích pohlavních orgánech člověka a šimpanze. Cíl práce byl splněn, mimo to byly identifikovány dvě dosud nepublikované sekvence, jejichž exprese je specifická pro samčí rostliny, a byl nalezen pohlavní dimorfismus na úrovni exprese ve vegetativních orgánech.

Abstract:

Proposed explanation for evident adaptive evolution of genes expressed in male reproductive organs of humans and chimpanzees is based on a genomic conflict resulting from germ cell competition during spermatogenesis. The aim of this work was to find out, whether analogous genes in Silene latifolia and Silene diclinis, where no comparable competition during microsporogenesis occurs, did evolve under positive selection since the divergence of these species as well. Two fifths of the analysed ESTs have recognisable homologues in the genome of Arabidopsis thaliana, but these homologues do not show anther specific expression. Most analysed ESTs have no homologues in A. thaliana. Their male specific function is apparently evolutionarily rather young. Expression analysis using RT-PCR revelaed that only 6 out of 40 ESTs for which anther-specific expression was published, are expressed specifically in anthers. Expression of 20 ESTs revealed sex-specific differences. S. latifolia and S. diclinis orthologues of 6 anther specific genes and 9 genes with distinct sex-specific expresion patterns were compared and no evidence for ongoing positive selection was found. On the contrary, sequences seem to be much more conserved than genes unrelated to male reproductive organs. Genes expressed in male reproductive organs of dioecious Silene species supposedly underwent adaptive evolution before S. latifolia and S. diclinis diverged, but since then they evolved very little or not at all, and currently are not under positive selection pressure. The genomic conflict resulting from germ cell competition during spermatogenesis thus remains the favourable explanation of positive selection of male reproductive organ specific genes in humans and chimpanzees. The appointed task was completed, in addition, two previously unknown ESTs showing male specific expression were identified and evidence for sexual dimorfism on the level of transcription in vegetative organs was found.
