The effect of alcohol on the dissolution behaviour of carrageenan based matrix tablets

Abstract:

Tato bakalářská práce se zabývá matricovými tabletami na bázi karagenanu a chitosanu s účinnou látkou tramadol hydrochlorid a působením alkoholu na uvolňování této účinné látky. Cílem práce bylo prostudovat vliv alkoholu na mechanismus a rychlost uvolňování léčiva ze studovaných matricových tablet a prokázat možný "alcohol-induced dose dumping" efekt. Metodou přímého lisování byly připraveny 4 formulace matricových tablet s obsahem 50 a 79 % komponent tvořících polyelektrolytový komplex (chitosan a karagenan) a s různým poměrem těchto komponent. S cílem prostudovat vliv alkoholu na tvorbu polyelektrolytového komplexu a následně na rychlost a mechanismus uvolňování modelového léčiva ze studovaných tablet byly provedeny disoluční testy v médiu bez alkoholu a v médiu s obsahem 40 obj. % alkoholu. Množství uvolněné účinné látky bylo stanoveno pomocí UV/VIS spektroskopie. Na základě získaných disolučních profilů byl vyhodnocen vliv přítomnosti alkoholu v disolučním médiu na mechanismus a rychlost uvolňování modelového léčiva tramadol hydrochloridu ze studovaných formulací.

Abstract:

This bachelor thesis deals with matrix tablets based on carrageenan and chitosan with the active substance tramadol hydrochloride and the action of alcohol on the release of this active substance. The aim of the study was to study the effect of alcohol on the mechanism and rate of drug release from the studied matrix tablets and to demonstrate the possible "alcohol-induced dose dumping" effect. 4 matrix tablet formulations containing 50 and 79 % of polyelectrolyte complex components (chitosan and carrageenan) and with different ratios of these components were prepared by direct compression. In order to study the effect of alcohol on the formation of the polyelectrolyte complex and subsequently on the rate and mechanism of release of the model drug from the studied tablets, dissolution tests were performed in a medium without alcohol and in a medium containing 40 % alcohol by volume. The amount of active substance released was determined by UV/VIS spectroscopy. Based on the obtained dissolution profiles, the effect of the presence of alcohol in the dissolution medium on the mechanism and rate of release of the model drug tramadol hydrochloride from the studied formulations was evaluated.
