Characterisation of the reaction of immobilised tyrosinase

Abstract:

Tato bakalářská práce se věnuje studiu aktivity enzymu tyrosinasy imobilizované na piezoelektrickém sensoru kovalentní vazbou technikou tvorby molekulových monovrstev a technikou zesíťování přes glutaraldehyd a studiem tvorby polymerního chinoidního filmu na povrchu imobilizovaného retikula v přítomnosti pyrokatecholu jako substrátu enzymu. Pro měření změn frekvence a odporu byl použit QCM metr a amperometrická detekce probíhala pomocí potenciostatu EmStat. Imobilizovaná vrstva na krystalech modifikovaných kovalentně navázanou tyrosinasou podléhala rychlé degradaci a aktivita tyrosinasy také rychle klesala. Na krystalech nebyla viditelná depozice chinoidních polymerů jak v případě QCM měření, tak v případě amperometrické detekce. Naopak u krystalů modifikovaných zesíťovanou tyrosinasou bylo hnědnutí imobilizované vrstvy viditelné. V případě QCM měření se hnědnutí retikula projevilo již po prvním měření a s každým dalším měřením byla tato vrstva tmavší, takže bylo prokázáno usazování chinoidních polymerů. Když byl k systému připojen potenciostat a na elektrodu přiveden záporný elektrický potenciál, chinoidní polymery se začaly viditelně usazovat až po několika měřeních a to jen v okrajových oblastech imobilizované vrstvy. Větší část chinoidních produktů byla zpětně redukována na pyrokatechol.

Abstract:

This bachelor's thesis is focused on study of activity of the enzyme tyrosinase immobilised on the piezoelectric sensor by covalent attachment using the techniques of self-assembled molecular monolayers and alternatively cross-linking via glutaraldehyde. Thus obtained biosensors were used to study the formation of quinone polymers on the surface of the immobilised reticulum in the presence of catechol as a substrate of the enzyme. For the measurement of frequency and resistance change the QCM meter was used and the potentiostat EmStat served for amperometric detection. The immobilised layer on the crystals modified by covalently attached tyrosinase undergone quick degradation and the activity of tyrosinase decreased quite quickly, too. There was visible deposition of quinone polymers neither in case of QCM measurement, nor in case of amperometric detection. On the other hand, on the crystals modified with cross-linked tyrosinase the browning of the immobilised layer was visible. In case of QCM measurement the browning of reticulum was visible immediately after the first measurement, therefore the deposition of quinone polymers was proved. When potentiostat was connected to the system and negative electric potential was applied to the electrode, quinone polymers were visibly depositing after several measurements and only in marginal areas of the immobilised layer. Larger amount of quinone products was reduced back to catechol.
