Neuroimmunological aspects of the development of pharmacoresistance in epilepsy

Abstract:

Hlavním tématem mé disertační práce je komplexní problematika neurozánětu a jeho podíl na etiologii epileptogeneze a vzniku farmakorezistence epilepsie. Hlavním cílem práce bylo identifikovat potenciální biomarkery epileptogeneze a farmakorezistence. V první práci (Aulická a Česká et al, Epilepsy Research, 2022) jsme analyzovali cytokinovo-chemokinové profily ve vzorcích hippocampů od pacientů s MTLE-HS versus post mortem zdravým kontrolám. Nejdůležitějším výsledkem první autorské práce je průkaz zvýšené exprese STAT3 u pacientů s HS. Specifické cytokiny a chemokiny by mohli v budoucnu představovat potenciální terapeutické cíle. Ve druhé práci (Česká K et al, Eur J Paediatr Neurol., 2023) jsme se zaměřili na analýzu specifických cytokinů a chemokinů ve vzorcích mozkomíšního moku a séra u dětských pacientů s farmakorezistentní a farmakodependentní epilepsií oproti zdravým kontrolám. Naše výsledky poukazují na možnost identifikace potenciálních biomarkerů farmakorezistence epilepsie u dětských pacientů.

Abstract:

The main topic of my doctoral thesis is the complex issue of neuroinflammation and its involvement in the etiology of epileptogenesis and the development of pharmacoresistant epilepsy. The main purpose of the work was to identify potential biomarkers of epileptogenesis and pharmacoresistance in epilepsy. In the first work (Aulická and Česká et al., Epilepsy Res, 2022) we analyzed cytokine-chemokine profiles in hippocampal samples from patients with MTLE-HS versus postmortem healthy controls. Most importantly, we found expressions of STAT3 in the HS patients. Specific cytokines & chemokines may represent therapeutic targets. In the second work (Česká et al., Eur J Paediatr Neurol, 2023) we focused on the analyses of specific cytokines & chemokines in cerebrospinal fluid and serum samples of pediatric patients with drug-resistant and drug-dependent epilepsy versus healthy controls. Our results indicate the possibility of identifying biomarkers of pharmacoresistance in pediatric patients.
