Genetic determinants in the etiopathogenesis of oral lichen planus

Abstract:

Orální lichen planus (OLP) je onemocnění sliznice dutiny ústní s nejasnou etiopatogenezí. Současná literatura naznačuje, že OLP je způsobeno poškozením bazálních keratinocytů způsobeným shlukem CD8+ T-buněk spouštějící apoptózu buněk orálního epitelu. Některá systémová onemocnění, jako je diabetes mellitus, hypertenze, ulcerativní kolitida aj. jsou s OLP spojovány. Cílem této práce bylo vypracovat literární rešerši vztahující se k onemocnění OLP z genetického a mikrobiologického hlediska. Cílem praktické části práce bylo imunohistochemické stanovení biomarkeru p53, izolace DNA z patologické tkáně sliznice dutiny ústní pacientů s OLP a zavedení metodiky sekvenování nové generace pro genetický screening pomocí komerčního panelu ke stanovení mutací ve vybraných onkogenech. Byla vypracovaná literární rešerše zaměřující se jednonukleotidové polymorfizmy cytokinů a onkogenů a jejich expresi, dále na osídlení dutiny ústní mikrobiotou u lidí trpící onemocněním OLP.

Abstract:

Oral lichen planus (OLP) is a disease of the oral mucosa with unclear etiopathogenesis. The current literature suggests that OLP is caused by damage to basal keratinocytes caused by a cluster of CD8 + T cells starting apoptosis of oral epithelial cells. Some systemic diseases, such as diabetes mellitus, hypertension, ulcerative colitis, etc., are associated with OLP. The aim of this work was to create a literature research apply of OLP disease from a genetic and microbiological perspective. The aim of the practical part of the work was to create immunohistochemical determination of biomarker p53, isolation of DNA from pathological oral mucosal tissue of patients with OLP and introduction of a new generation sequencing methodology for genetic screening with the use of a commercial panel to determine mutations in selected oncogenes. A literature research was made focusing a literature search focusing on single nucleotide polymorphisms of cytokines, oncogenes, their expression, and next the colonization of the oral cavity by microbiome in people suffering from OLP disease.
