Synthesis of thioamide analogs of known "K" oxime reactivators of cholinesterases

Abstract:

Tato bakalářská práce je zaměřena na syntézu nových biskvartérních pyridinaldoximových sloučenin jako potenciálních reaktivátorů inhibovaných cholinesteras. Strukturně jsou tyto sloučeniny odvozeny od oximů K027, K048 a K203, které poskytly slibné výsledky v oblasti reaktivace organofosfáty inhibovaných cholinesteras. Konkrétně se jedná o substituci amidové skupiny za thioamidovou. Teoretická část je zaměřena na charakteristiku organofosfátů a cholinesteras, je zde popsán inhibiční účinek nervově paralytických látek na cholinesterasy a možnosti případné terapie otrav těmito látkami. Dále jsou zde shrnuty poznatky od paramteru pKa a o stabilitě sloučenin. Praktická část práce se zabývá syntézou jednotlivých biskvartérních solí a měřením hodnot pKa a stability těchto sloučenin. Tyto výsledky jsou dále porovnány s analogickými „K-oximy“ K027, K048 a K203.

Abstract:

This bachelor thesis is focused on the synthesis of novel bisquaternary pyridinaldoxime compounds as potential reactivators of inhibited cholinesterases. Structurally, these compounds are derived from the oximes K027, K048 and K203, which have provided promising results in the reactivation of organophosphate-inhibited cholinesterases. Specifically, they involve the substitution of an amide group for a thioamide group. The theoretical part focuses on the characterization of organophosphates and cholinesterases, the inhibitory effect of nerve paralytic agents on cholinesterases and the possibilities of possible therapy of poisoning by these agents. Furthermore, the findings from the pKa parameter and the stability of the compounds are summarized. The practical part of the thesis deals with the synthesis of individual bisquaternary salts and measurement of pKa values and stability of these compounds. These results are further compared with the analogous "K-oxime" K027, K048 and K203.
