Význam lektinů patogenních mikroorganismů se zaměřením na lektiny u rodu Candida: průkaz a charakteristika lektinů Candida albicans

Abstract:

Lektiny patogenních mikroorganismů hrají významnou roli ve složitých interakcích mezi patogeny a hostitelskými buňkami. Přestože primárně fungují jako adhezivní molekuly, účastní se i pochodů na adhezi navazujících. Díky svým vlastnostem nacházejí široké uplatnění v nejrůznějších vědních oborech. Adheze patogenních mikroorganismů na tkáně hostitele je nezbytná pro rozvoj většiny infekčních onemocnění. Možnost léčby založená na přerušení této interakce nabývá v souvislosti se vznikem patogenních kmenů rezistentních k antibiotikům stále většího významu. Pro inhibici adheze je však nutné důkladně poznat adhezivní molekuly, tudíž i lektiny. Podmíněně patogenní kvasinkovité organismy rodu Candida způsobují nejvíce mykózních onemocnění u lidí, přičemž v naprosté většině případů je původcem onemocnění Candida albicans. Přes svůj význam nejsou lektiny Candida albicans ani zdaleka prostudovány. Candida albicans dokáže adherovat na mnohé povrchy a při některých těchto interakcích se prokazatelně uplatňují lektiny, ale žádný z nich dosud nebyl přesně popsán. Situaci komplikují kmenové rozdíly ve specifitě adhezinů a pozorovaný vliv kultivačních podmínek na jejich expresi, což způsobuje časté rozpory v publikovaných výsledcích. Teoretická část této práce obsahuje shrnutí funkcí lektinů patogenních mikroorganismů a mapuje dosavadní pokroky ve výzkumu lektinů Candida albicans a dalších významných zástupců rodu Candida. Smyslem praktické části je průkaz a charakteristika lektinů Candida albicans.

Abstract:

Lectins are important in both symbiotic and pathogen interaction between some microorganisms and hosts because microbial lectins may play an essential role in mediating adhesion to cells and mucosal surfaces of the host. Adhesion protects pathogens from natural cleansing mechanisms (airflow in the respiratory tract, urine flow in the urinary tract), provides the pathogens with better access to sources of nutrition and supports the penetration of pathogens into the tissues. Candida albicans (dimorphic yeast) is the most important human fungal pathogen. Attachment of the yeast form to host cells is believed to be the first and crucial step in the pathogenesis of Candida infections. Two Candida albicans lectins have been partially characterized (fucoside-binding cell wall mannoprotein, fimbrial adhesin) but this fungus appears to use more than two lectin-like adhesins. In this diploma thesis there was shown that Candida albicans can also bind to mannan and a mannan-binding protein was isolated and described. This putative lectin was found in the strain Candida albicans 8186. Protein has a molecular mass of 12,0 kDa and binding of mannan is promoted by Ca2+. The N-terminal part of mannan-binding protein is the same as the mannose-binding lectin from Ralstonia solanacearum and resembles the fucose-binding lectin from Pseudomonas aeruginosa. The gene for mannan-binding protein was amplified and transformed into Escherichia coli. Preparation of this genetically modified organism allows avoiding the work with pathogenic fungus and will facilitate the future study of the mannan-binding protein.
