Biodegradable polymers-based nanomaterials applied for drug delivery system

Abstract:

Bakalářská práce se zabývá nanomateriály z biodegradabilních polymerů s potenciálem pro využití v cílené distribuci léčiv. Teoretická část shrnuje základní vlastnosti, možnosti přípravy a využití některých polymerů s vysokým potenciálem pro kontrolované podání terapeutických látek. Dále se věnuje formám, v jakých se tyto systémy aplikují do těla pacienta. V experimentální části je popsán postup přípravy nanočástic z kyseliny hyaluronové, na které byl navázán biotin jako bioaktivní molekula. Součástí práce je také optimalizace postupu jeho kvantifikace, která byla prováděna spektrofotometricky. U připravených částic modifikovaných biotinem byla ověřena jeho skladovací stabilita. Byla také testována odolnost vazby biotinu na nanočásticích, a to po působení silně kyselého a zásaditého prostředí jako simulace extrémních podmínek v organismu.

Abstract:

This bachelor thesis deals with nanomaterials made of biodegradable polymers with potential for use in targeted drug delivery. The theoretical part summarizes general properties, methods of preparation and specific application areas of several polymers with high potential for controlled delivery of therapeutic substances. It also focuses on the way how these systems get into the patient's body. The experimental part describes the process of preparation of nanoparticles of hyaluronic acid to which biotin has been bound as a bioactive molecule. The thesis also includes the optimization of the process of its quantification, which was measured spectrophotometrically. The prepared biotin-modified particles have been tested for storage stability. Finally, the resistance of biotin binding to nanoparticles has also been tested in acidic and alkaline conditions as a simulation of extreme conditions in the body.
