Abstract:

Celý obsah předložené diplomové práce je zaměřen na problematiku benzodiazepinových a pyrrolobenzodiazepinových sloučenin. I přesto, že od jejich objevení uplynulo více než 50 let, představuje tato oblast stále velmi živé a zajímavé odvětví nejen z pohledu organické chemie, ale i farmacie. Rešeršní část současně shrnuje jak využití a praktické aplikace těchto významných bioaktivních látek, tak i jejich publikované metody syntéz využívané k jejich přípravě. Experimentální část představuje hned několik jednoduchých přístupů vedoucích k syntéze benzodiazepinů a pyrrolobenzodiazepinů ze snadno dostupných a laciných výchozích látek. Práce zpočátku popisuje optimalizaci podmínek přípravy 3-(3-hydroxypropylamino)chinolin-2,4(1H,3H)-dionů. Tyto vhodné prekursory, po vystavení účinkům thionylchloridu, tetramethylguanidinu a hydridu sodného v různém pořadí, poskytly benzo[e][1,4]diazepin-2,5-diony a benzo[e]pyrrolo[1,2-a][1,4]diazepin-5,11-diony. Průběhy a výsledky reakcí jsou podpořeny výsledky analytických instrumentálních metod, jejichž spektra jsou komentována ve výsledkové a diskusní části.

Abstract:

The entire content of the submitted thesis is focused on the problems of benzodiazepine and pyrrolobenzodiazepine compounds. Despite the fact that more than 50 years have passed since their discovery, the area is still a very current and interesting industry not only in terms of organic chemistry but also pharmacy. At the same time, the research part summarizes both the use and practical application of these important bioactive substances and their published methods of synthesis used for their preparation. The experimental part presents several simple approaches leading to the synthesis of benzodiazepines and pyrrolobenzodiazepines from readily available and cheap starting materials. The work initially describes the optimization of the preparation conditions of 3-(3-hydroxypropylamino)quinoline-2,4(1H,3H)-dione. After exposure to thionyl chloride, tetramethylguanidine and sodium hydride in different order, these suitable precursors yielded benzo[e][1,4]diazepine-2,5-diones and benzo[e]pyrrolo[1,2-a][1,4]diazepine-5,11-diones. The course and results of the reactions are supported by the results of analytical instrumental methods, whose spectra are commented on in the results and discussion sections.
