General aspects of N-glycosylation in cancer

Abstract:

Tato bakalářská práce má za cíl stručně shrnout poznatky o N-glykosylaci a jejich změnách během maligní transformace, které hrají roli v následné zvýšené invazivitě nádoru. V první části této práce jsou stručně shrnuty nejčastější posttranslační modifikace proteinů a následně popsán proces N-glykosylace. Dále jsou popsány samotné změny v N-glykosylaci proteinů a jejich následky s důrazem na pozměněné větvení bočních řetězců N-glykanů a tvorbu Lewisových antigenů. Poslední část je věnována deregulaci glykosylace jako nádorovému biomarkeru a nádorové terapii.

Abstract:

This bachelor theses shortly summarises knowledge about N-glycosylation a nad its chnges in malignant transformation which plays key role in tunor invasivity. First part of the thesis focus on the most common posttranslational modifications of proteins and then describes process of N-glycosylation. Moreover, there are described changes in proces of N-glycosylation with emphasis on aberrant branching of N-glycans and synthesis of Lewis antigens. Last chapter focuses on deregulation of glycosylation as cancer biomarkers and cancer therapy.
