Rho signaling system and its role in cytoskeletal remodelling and cell

Abstract:

Signální dráha rodiny Rho se podílí na regulaci aktinového cytoskeletu a migrace buněk. Ovlivňuje nejen vznik pohybových orgánů a adhezivních struktur, ale řídí také buněčnou polaritu, cyklus a dynamiku aktinu. Tato bakalářská práce popisuje jednotlivé komponenty signální dráhy Rho se zaměřením na její nejznámější a nejlépe prozkoumané členy: Rac1, RhoA a Cdc42. Pod vlivem malé GTPázy Rac1 vznikají lamelipodia, GTPáza RhoA reguluje formování stresových vláken a GTPáza Cdc42 řídí vývoj filopodií.

Abstract:

The members of the Rho GTPase family have been shown to regulate many aspects of intracellular actin dynamics and cell motility.The Rho signaling pathway not only regulates the development of migrating structures and adhesion sites, it also coordinates cell polarization and organisation of the actin cytoskeleton. This thesis describes individual components of the signaling pathway with emphasis on its most significant members: Rac1, RhoA, Cdc42. Activation of the small GTPase Rac1 leads to lamellipodia formation, RhoA expression encourages stress fiber formation and activity of the small GTPase Cdc42 enhances formation of filopodia.
