Analysis of hyaluronic acid using liquid phase separation techniques

Abstract:

V této diplomové práci je optimalizováno kvantitativní stanovení kyseliny hyaluronové ve farmaceutických výrobcích pomocí kapalinové chromatografie ve spojení s hmotnostní spektrometrií či fluorescenční detekcí. Jsou podrobně popsány hyaluronidázy, jejich typy a mechanismus štěpení. Fragmentace byla provedena enzymem bakteriální hyaluronanlyáza a účinnost ověřena spektrofotometricky. Práce se zaměřuje na derivatizaci hyaluronanových fragmentů pomocí 2-aminoakridonu a jejich následnou chromatografickou separaci v systémech s obrácenými fázemi. V práci jsou uvedeny další chromatografické, elektroforetické a spektrální metody ke stanovení hyaluronanu. Zahrnuto je i aktuální využití, výroba a unikátní vlastnosti této látky.

Abstract:

In this thesis, the quantification of hyaluronic acid in pharmaceutical formulations using liquid chromatography with mass spectrometry or fluorescent detection is optimized. Different types and cleavage mechanism of the hyaluronidases is described. The fragmentation was performed by the bacterial hyaluronan lyase and cleavage was confirmed with spectrophotometry. The thesis is focused on derivatization techniques of hyaluronan fragments with 2-aminoacridone followed by reversed phase chromatography. The spectral, chromatographic and electrophoretic methods developed for hyaluronan determination are introduced. Also current applications, production and unique characteristics of this substance are included.
