Effect of cobalt on growth and total vitamin B12 content of Arthrospira maxima and consortium with microalgae

Abstract:

V úvodních kapitolách se tato práce zabývá literárním shrnutím studovaných sinic, mikrořas a vitaminu B12. V experimentální části práce byla optimalizována mikrobiální metoda stanovení vitaminu B12, pomocí bakterie Lactobacillus delbrueckii subsp. lactis. Dále byly v experimentální části testovány vhodná kultivační média pro konsorcium sinice Arthrospiry maxima a mikrořasy Chlorelly vulgaris. Protože je kobalt součástí struktury vitaminu B12, byl studován jeho vliv na růst a obsah vitaminu B12 v biomase. Hodnocení růstu bylo stanoveno spektrofotometricky a váhovým přírůstkem. Signifikantní rozdíl v obsahu vitaminu B12 byl naměřen u konsorcia kultivovaného ve Spirulina médiu s 10 µM hexahydrátu dusičnanu kobaltnatého. Mezi Arthrospirou maxima kultivovanou v kontrolním Spirulina médiu a médiu s přídavkem kobaltu nebyl naměřen statisticky významný rozdíl. Odlišný obsah vitaminu B12 nebyl stanoven ani mezi Arthrospirou maxima a konsorciem v kontrolním médiu. Kobalt také neměl vliv na optickou denzitu a váhový přírůstek Arthrorpiry maxima a konsorcia.

Abstract:

In the preliminary, thesis deals with a literature summary of the studied cyanobacteria, microalgae and vitamin B12. In the experimental part, the microbial method for the determination of vitamin B12 was optimized, using the bacterium Lactobacillus delbrueckii subsp. lactis. Furthermore, suitable culture media for the consortium of cyanobacteria Arthrospira maxima and microalgae Chlorella vulgaris were tested in the experimental part. As cobalt is part of the structure of vitamin B12, its effect on growth and vitamin B12 content in biomass was studied. Growth evaluation was determined spectrophotometrically and by weight gain. A significant difference in vitamin B12 content was measured in a consortium grown in Spirulina medium with 10 µM cobalt nitrate hexahydrate. No statistically significant difference was measured between Arthrospira maxima cultured in Spirulina medium and cobalt-added medium. A different content of vitamin B12 was not determined between Arthrospira maxima and the consortium in the control medium. Cobalt had no effect towards optical density or weight gain of the Arthrorpira maxima and the consortium.
