Capillary electrophoresis as a member of the metabolomic analytical toolbox

Abstract:

Kapilární elektroforéza je analytická technika, která je založena na separaci iontů v elektrickém poli. Tato metoda je velice oblíbená, zejména proto, že pro analýzu vyžaduje malé objemy vzorků, chemikálií, a tudíž produkuje minimum odpadů. Další přednosti této metody jsou její selektivita a snadná automatizace. Pro své výhody je velice často používaná v metabolomických studií. Metabolomika je vědní disciplína, která studuje bilanci nízkomolekulárních látek uvnitř i vně buňky. Nachází uplatnění v hledání nových markerů, které mohou sloužit k diagnostickým účelům. Asistovaná reprodukce vyžaduje diagnostiku embrya k posouzení jeho kvality a implantačního potenciálu. Současným trendem je vyvinutí neinvazivních technik, které budou poskytovat nové informace o fyziologickém stavu embrya a zlepšit tak jeho výběr. Předmětem diplomové práce je aplikovat kapilární elektroforézu pro analýzu kultivačních médií používaných v asistované reprodukci. Práce je zaměřena na analýzu aminokyselin, které jsou součástí kultivačního média a jejichž bilance může odrážet fenotyp embrya. Pro detekci aminokyselin byla použita bezkontaktní vodivostní detekce.

Abstract:

Capillary electrophoresis is an analytical technique based on the separation of ions in an electric field. This technique is very attractive for the low sample consumption and negligible amounts of waste produced. There are other advantages like selectivity and easy automation. For its advantages, it is widely used technique in metabolomic studies. Metabolomics is a scientific field, which is focused on the measurement of the low molecular weight compounds inside and outside the cell. It is employed for seeking of novel markers which can serve for diagnostic purposes. Assisted reproduction requires diagnosis of an embryo for the evaluation of its quality and implantation potential. Contemporary goal is to develop non-invasive techniques, which will provide new information about the physiological state of the embryo thus improve embryo selection. The subject of this thesis is to apply capillary electrophoresis for an analysis of culture media used in the assisted reproduction. This work is aimed at amino acids analysis presented in the common culture media. Amino acids turn-over can reflect phenotype of the embryo. For amino acids detection there was used contactless conductivity detection.
