Enantioselective catalysts containing thiourea scaffold

Abstract:

V rámci bakalářské práce byla provedena literární rešerše, týkající se enantiokatalytických vlastností chirálních thiomočovin. Na základě této rešerše byly navrženy struktury dvou nových thioacyl thiomočovin odvozených od (R)-1-fenylethanaminu a (1S,2S)-cyklohexan-1,2-diaminu. Úspěšně byla realizována syntéza (R)-1-(fenylkarbonthioyl)-3-(1-fenylethyl)thiomočoviny, která byla charakterizována NMR spektry a následně testována v modelové Henryho reakci 2-methoxybenzaldehydu s nitromethanem. Bylo zjištěno, že za přítomnosti uvedené thiomočoviny a octanu měďnatého reakce vůbec neprobíhá. Dále byla testována enantiokatalytická aktivita připravené thiomočoviny při redukci acetofenonu. Ani v tomto případě však nebyla požadovaná aktivita pozorována. Proto byl učiněn pokus o syntézu (1S,2S)-1-[(2-aminocyklohexyl)-3-fenylkarbonthioylthiomočoviny analogickým postupem. Bylo zjištěno, že tento postup není pro přípravu vhodný - vznikala směs obtížně separovatelných produktů.

Abstract:

Within the framework of this bachelor's work thorough literature retrieval was made regarding the enantiocatalytic properties of chiral thioureas. On the basis of this literature retrieval two new thioacyl thioureas were suggested which were derived from (R)-1-phenylethanamine and (1S,2S)-cyclohexan-1,2-diamine. The synthesis and NMR characterization of (R)-1-(phenylcarbonothioyl)-3-(1-phenylethyl)thiourea was successfully carried out and this thiourea was tested in model Henry reaction of 2-methoxybenzaldehyde with nitromethane. It was found that the reaction does not take place in the presence of above-mentioned thiourea and copper(II) acetate. Next the enatiocatalytic properties of prepared thiourea were tested in acetophenone reduction. However no enantioselectivity was observed. Therefore an attempt at synthesis of (1S,2S)-1-[(2-aminocyclohexyl)-3-phenylcarbonothioylthiourea was carried out. Unfortunately it was found that an analogous procedure which was successfully used for synthesis of (R)-1-(phenylcarbonothioyl)-3-(1-phenylethyl)thiourea is impossible due to occurrence of non-separable by-products.
