HPCL evaluation of quaternary benzophenanthridine alkaloids in selected species of the family Papaveraceae

Abstract:

Kvartérní benzofenanthridinové alkaloidy (KBA) jsou malou skupinkou isochinolinových alka-loidů a jsou biosyntetizovány z fenylalaninu. Jejich hlavním zdrojem je čeleď Papaveraceae, v malém množství se také vyskytují v čeledi Fumariaceae, Rutaceae a Ranunculaceae. Nejznámějšími benzofenanthridiny jsou sanguinarin a chelerythrin. Jsou nazýváné jako hlavní alkaloidy a vykazují velmi široké spektrum biologických účinků např.: protizánětlivý, antimikrobiální nebo antifungální účinek.Z řady studií je známo, že v rostlinách jsou hlavní KBA velmi často doprovázeny dalšími benzofenathridiny, které mají stejnou strukturu ale jinak substituované jádro. Jedná se o sanguirubin, sanguilutin, chelilutin, chelirubin a makarpin. Nazývají se minoritními alkaloidy, protože se vyskytují ve velmi malém množství. Z důvodu omezené dostupnosti jsou doposud informace o jejich biologických účincích velmi limitované Tato práce byla vypracována jako součást systematického studia kvartérních ben-zo[c]fenanthridinových alkaloidů a byla zaměřena na získání většího množství informací o mino-ritních kvartérních alkaloidech. Cílem této práce proto bylo zjistit kvalitativní a kvantitativní zastoupení KBA v rostlinných druzích, které byly dosud popsány jako jejich zdroje a jejich změny v průběhu vegetačního období pomocí metody HPLC. V návaznosti na tyto poznatky pak byly formulovány závěry o nejvhodnějších zdrojích pro izolaci majoritních i minoritních alkaloidů. Pomocí vytvořené HPLC metody jsme stanovili obsah benzofenathridinů v šesti druzích čeledi Papaveraceae a u třech z nich jsme zhodnotili změny v obsahu alkaloidů v průběhu vegetace. Pozorované změny jsou v souladu s obecnýmí poznatky o biosyntéze alkaloidů. Nejvhodnějším zdrojem hlavních alkaloidů se nám jeví Dicranostigma lactucoides a nejvhodnějším zdrojem minoritních alkaloidů s výjimkou makarpinu Sanguinaria canadensis. A co víc, pouze v S. canadensis jsme identifikovali sanguilutin, v ostatních zdrojích nebyl stanoven. Potenciálními zdroji makarpinu jsou kořeny pouze dvou rostlinných druhů – S. lasiocarpum a M. microcarpa.

Abstract:

The benzophenanthridine alkaloids (QBA) are a small group of isoquinoline alkaloids and are biosynthetised from phenylalanine. These alkaloids mostly occur in families Papaveraceae, Fu-mariaceae, and Rutaceae. The best known QBA are sanguinarine and chelerythrine. These so called main benzophenanthridine alkaloids show a wide spectrum of biological activities, such as antiinflamatory, antimicrobacterial, or antifungal. The main quaternary benzophenanthridines are in plants very often accompanied by benzophenanthridines with the same skeleton but different substitution. These are sanguirubine, sanguilutine, chelilutine, chelirubine, and, rarely, macarpi-ne. These alkaloids are called minor alkaloids, because their occur in very small amounts. The knowledge about their biological activities is very limited due to their poor availability. This work was elaborated as a one part of systematic study of benzophenanthridine alkaloids and was focused on obtaining larger amount of information about the minor benzophenanthridine alkaloids. The aim of this work was to determine the occurence and quantity of benzophenanthri-dine alkaloids in herbal species that have been described as their main sources and assess changes in their content during the vegetation peroid using the HPLC method. For the quantitative and qualitative determination of benzophenanthridine alkaloids, the HPLC method was established. Subsequently, the most suitable sources of main and minor QBA were selected. Three types of stacionary phases were tested and the best results were obtained with a reversed-phase column SynergiTM MAX-RP. We determined and quantified benzophenanthridine alkaloids in six species of the family Papa-veraceae and in three of them the seasonal changes in the QBA content were assessed. The ob-served changes in the QBA content were in agreement with a general knowledge about their bi-osynthesis. Dicranostigma lactucoides was found as the most suitable source for isolation of the main alkaloids and Sanguinaria canadensis for minor alkaloids except macarpine. Moreover, S. canadensis is the only source of sanguilutine from species that we studied. The best sources of macarpine are two species – Stylophorum lasiocarpum and Macleaya microcarpa.
