Quantitative anatomy of xylem in species Vitis vinifera and Clematis vitalba

Abstract:

V této bakalářské práci se věnuji srovnání některých strukturních znaků xylému u rostlin Vitis vinifera a Clematis vitalba. Strukturní znaky xylému určují rychlost a efektivitu transportu vody a v ní rozpuštěných látek do celého rostlinného těla. Cílem práce bylo porovnat délku cév a jejich průměr u obou rostlinných druhů vzhledem k jejich příslušnosti k liánám a také zjistit, zda se vnitřní průměr cév v jejich podélné ose mění, případně jak moc a zda může mít tento znak vliv na rychlost transportu xylémem. Pro identifikaci jednotlivých cév byla použita metoda nástřiku silikonu s fluorescenčním barvivem. U obou druhů jsem pozorovala znaky typické pro liány jako jsou dlouhé cévy (nejdelší naměřené cévy byly u Vitis 48 cm, u Clematis 56 cm) a velký vnitřní průměr cév (největší vnitřní průměr naměřený u Vitis byl 110 µm, u Clematis 133 µm). Také jsem zjistila, že se vnitřní průměr cév v jejich podélné ose velmi výrazně mění. Největší variability dosahovaly dlouhé cévy a v některých případech docházelo ke změně vnitřního průměru i o více než 100 % (za 100 % je považována hodnota průměrného vnitřního průměru dané cévy). Byl proto vypočítán teoretický vliv variability vnitřního průměru na odpor cévy. Ukázalo se, že s rostoucí mírou variability exponenciálně roste odpor cévy. Nejvyšší nárůst odporu (až 3x oproti ideální cévě s neměnným vnitřním průměrem) byl vypočítán u nejdelších cév s velkým vnitřním průměrem, které tvoří pouze malou část všech cév. Tyto cévy se však významně podílejí na celkové hydraulické vodivosti xylému a změny odporu v této skupině cév jsou tedy z pohledu celkové rychlosti transportu ve stonku významné.

Abstract:

In this thesis I compared structure of xylem in species Vitis vinifera and Clematis vitalba. Structure of xylem determines rate and efficiency of water transport. First aim of this thesis was to compare vessel length and vessel diameter in both liana species. Second aim was to determine a variation of vessel inner diameter along vessel longitudal axis and quantify its effect on xylem transport. The silicone injection method was used for identification of individual vessels. Long (the longest vessel in Vitis was 48 cm, in Clematis 56 cm) and wide (the widest vessel diameter in Vitis was 110 µm, in Clematis 133 µm) were observed in both plant species. I also found out that vessel diameter was very variable along its longitudinal axes. The variability of vessel inner diameter was the highest in long and wide vessels. In some cases change of their diameters was more than 100 % (100 % is mean vessel diameter calculated for each vessel). Therefore I determined theoretical influence of vessel diameter variance on vessel resistance. I foud that vessel resistance increased exponentially with increasing variance of diameter. The highest increase of vessel resistance (up to 3x compared to ideal vessel with constant diameter) was determined for long vessels with wide diameters. Although there is generally a small number of long and wide vessels in a typical liana stem they contribute significantly to total hydraulic conductivity. Such considerable increase of vessel resistance in this size class can significantly affect xylem transport at the whole plant level.
