Wheat seed vigour, its heritability and potential use to reduce the effects of drought

Abstract:

K nejčastějším abiotickým stresorům působící na klíčivost semen a vývoj rostlin se řadí sucho a chlad. Proto provenience a potenciál genotypů pro snížení dopadů stresu má zásadní význam. Perspektivní metody hodnotící vitalitu semen mají nezastupitelnou úlohu nejen v zahraničí. Interakce genotypu a prostředí ve vztahu k abiotickým stresům byla hodnocena u pšenice seté ozimé (Triticum aestivum L.) v letech 2018 až 2020. Na podzim roku 2018 bylo zaseto 6 genotypů pšenice v areálu Mendelovy univerzity v Brně do osmi plastových nádob. Od dubna roku 2019 byly nádoby udržovány pod různými vláhovými režimy a zároveň byla 3x za vegetaci hodnocena velikost kořenového systému pomocí elektrické kapacity. Elektrická kapacita kořenů byla měřena v růstové fázi BBCH 36, v BBCH 55 a v BBCH 71. Semena těchto šesti genotypů byla testována při 10 °C a −5 barech, za využití polyethylenglykolu. Vitalitu představoval počet vyklíčených semen po 7 a 14 dnech a parametry z digitální analýzy obrazu: délka, plocha, průměr a objem kořenů a plumuly. Následně byl hodnocen vliv genotypu a vláhového režimu s velikostí kořenového systému (VKS) na hmotnost semen / rostlinu, na HTS a na vitalitu semen. Nakonec byla vyhodnocena dědivost vitality. Statisticky průkazný vliv na hladině významnosti p ≤ 0,05 byl zjištěn u hmotností semen na rostlinu (HSR) v závislosti na genotypu. Genotypy s největšími hmotnostmi zrn dosahovaly zhruba 5x vyšší hmotnosti než genotypy s nízkými hmotnostmi semen na rostlinu. Statisticky průkazné výsledky byly také zjištěny pro některé HTS genotypů v závislosti na variantě zavlažování. Statisticky průkazná závislost byla zjištěna mezi některými: HSR a VKS, VKS a vitalitou a variantami a vitalitou. Selekce rostlin na vysokou vitalitu byla neúspěšná, avšak na vitalitu nízkou, byla úspěšná. Pokud jako znak vitality bereme délku kořenů a plumuly, tak fenotyp se na proměnlivosti nízké hodnoty znaku v námi sledovaném souboru rostlin podílí ze 40 %.

Abstract:

The most common abiotic stressors affecting seed germination and plant development are drought and cold. Therefore provenance and potential of genotypes to reduce the effects of stress is crucial. The prospective methods for evaluating the vitality of seeds have an irreplaceable role not only abroad. Genotype-environment interaction in relation to abiotic stresses was evaluated in winter wheat (Triticum aestivum L.) from 2018 to 2020. In autumn 2018, 6 wheat genotypes were sown in eight plastic containers in the grounds of Mendel University in Brno. Since April 2019, the containers have been kept under different moisture regimes and alongside, the root system size was evaluated three times during vegetation using electrical capacity. The electrical capacity of the roots was measured in the growth phase BBCH 36, BBCH 55 and BBCH 71. Seeds of these six genotypes were tested at 10 °C and −5 bars, using polyethylene glycol. Seed vigour presented by the number of germinated seeds after 7 and 14 days and parameters from digital image analysis: length, area, diameter and volume of roots and plumules. Subsequently, the effect of genotype and moisture regime with root system size (VKS) on seed / plant weight, on weight of thousand seeds (HTS) and on grain vigour was evaluated. Finally, the inheritance of vitality was evaluated.A statistically significant effect on the level of significance p ≤ 0.05 was found for plant seed weights (HSR) depending on the genotype. The genotypes with the largest grain weights were about 5 times higher than the genotypes with low seed weights per plant. Statistically significant results were also found for some HTS genotypes depending on the irrigation variant. Statistically significant dependence was found between some: HSR and VKS, VKS and seed vigour, moisture régime and seed vigour. Selection of plants for high vitality was unsuccessful, but for low vitality, it was successful. If we take the length of roots and plumules as a sign of seed vigour, then the phenotype contributes to the variability of the low value of the sign from 40 %.
