Transcriptome changes induced by stress factors in tomato plants

Abstract:

V této diplomové práci se věnujeme zkoumání vlivu stresových faktorů na změny transkriptomu rajčete. Dva druhy rostlin rajčete, Solanum lycopersicum cv. Amateur a Solanum habrochites, byly ošetřeny oligandrinem, kyselinou beta-aminomáselnou (BABA) nebo inokulovány patogenem Oidium neolycopersici. Následně byly metodou qPCR sledovány změny v transkriptech PR (pathogenesis related) genů hrajících klíčovou úlohu při procesu rezistence rostlin vůči biotickým a abiotickým stresovým faktorům. Konkrétně byly sledovány geny patřící mezi defensiny (def), germin-like proteiny (germ), beta-1,3-glucanasy (glu), beta-1,4- glucanasy (gluco), heveiny (hev), chitinasy (chit), osmotiny (osm), systeminy (syst) a PR-1 proteiny. Po ošetření rostlin kyselinou beta-aminomáselnou a oligandrinem byly provedeny fytopatologické testy, které sledovaly, jaký vliv mají tyto látky na indukci rezistence proti patogenům Phytophthora parasitica a Botrytis cinerea. Susceptibilní kultivar rajčete Solanum lycopersicum cv. Amateur, náchylný na infekci patogenem Oidium neolycopersici, reagoval na všechna tři ošetření změnou v koncetraci sledovaných transkriptů. Po ošetření BABA navíc vznikla signifikantní protekce proti patogenu Phytophthora parasitica. Rezistentní kultivar rajčete, Solanum habrochaites, vykazoval změny v transkriptech po ošetření oligandrinem a inokulaci patogenem Oidium neolycopersici. Po ošetření BABA došlo pouze ke změně v transkriptů genů gluco3, P4 a P6. Tento kultivar vykazuje vysokou rezistenci k patogenům Oidium neolycopersici, Phytophthora parasitica a Botrytis cinerea i bez jakéhokoli ošetření a vliv BABA a oligandrinu na protekci nelze tedy jednoznačně určit. Po ošetření oligandrinem nedocházelo k signifikantnímu vlivu na výslednou rezistenci, ale docházelo ke zpomalení klíčení spor u patogenu Oidium neolycopersici.

Abstract:

In this thesis we study the transcriptome changes induced by stress factors in tomato plants. Two types of tomato plants, Solanum lycopersicum cv. Amateur and Solanum habrochaites, were treated by oligandrin, beta-aminobutyric acid (BABA) or inoculated by pathogen Oidium neolycopersici. Subsequently changes in transcripts of PR (pathogenesis related) proteins were monitored, that are important for defence reaction of plant against biotic and abiotic stress. By qPCR method the changes of transcripts for defensins (def), germins (germ), beta-1,3-glucanases (glu), beta-1,4-glucanases (gluco), heveins (hev), chitinases (chit), osmotins (osm), systemins (syst) and PR-1 proteins were monitored. Phytoprotection tests after treatment by BABA and oligandrin were done. These tests observed the effect of treatment on protection against Phytophthora parasitica and Botrytis cinerea. Solanum lycopersicum cv. Amateur, which is susceptible to Oidium neolycopersici, responded to all three inducers by changes in concentration of measured transcripts. Moreover, there was significant protection against Phytophthora parasitica after treatment by BABA. Solanum habrochaites, which is resistant to Oidium neolycopersici, showed changes in concentration of monitored transcripts after treatment by oligandrin and after inoculation with Oidium neolycopersici. After BABA treatment only transcripts of genes gluco3, P4 and P6 were significantly up-regulated. This type of tomato is highly resistant to Oidium neolycopersici, Phytophthora parasitica and Botrytis cinerea without any treatment and due to its high level of basal resistance no changes of protection can be clearly identified. After treatment by oligandrin there was no significant protection against Phytophthora parasitica but there was retardation of germination process of Oidium neolycopersici.
