Effect of selected substances affecting the central nervous system on metabolism of the endocannabinoids

Abstract:

Endokanabinoidy vykazují některé společné farmakologické vlastnosti s antidepresivy. Mají vliv na zpětné vychytávání monoaminů, čímž se řadí mezi potencionální terapeutické nástroje při léčbě depresivních a úzkostných poruch. Endokanabinoidní signalizace je klíčovým prvkem pro regulaci nálady a emocí. Jejím poškozením dochází k rozvoji deprese. Zvýšení endokanabinoidní signalizace by mohlo přispět ke stejnému terapeutickému účinku, kterým disponují současná antidepresiva. Cílem práce je přispět k rozšíření poznatků o modulaci endokanabinoidní signalizace pomocí antidepresiv a ověřit účinnost vybraných antidepresiv na zvýšení endokanabinoidních látek, anandamidu a 2-arachidonoylglycerolu, ve vybraných částech mozkové tkáně. Vybraná antidepresiva mirtazapin, trazodon a venlafaxin byla přidávána do standardní laboratorní diety potkanům po dobu 12ti týdnů. Odebrané vzorky mozkových tkání laboratorních potkanů byly rozděleny na jednotlivé části: levou hemisféru, pravou hemisféru, hypothalamus, mozkovou kůru a mozeček. Takto rozdělené části byly homogenizovány, extrahovány a následně byly vzorky měřeny metodou kapalinové chromatografie s hmotnostní spektrometrií. Bylo prokázáno, že mirtazapin statisticky významně snižuje koncentraci anandamidu v hypothalamu. Zatímco trazodon významně zvyšuje koncentraci anandamidu v pravé hemisféře mozkové tkáně, mozkové kůře a mozečku. Po podání venlafaxinu byla zaznamenána zvýšená koncentrace anandamidu v hypothalamu, mozkové kůře a mozečku. Koncentrace 2-arachidonoylglycerolu byla významně zvýšena po podání trazodonu v levé hemisféře, pravé hemisféře, hypothalamu, mozkové kůře a mozečku. Venlafaxin významně zvýšil koncentraci 2-arachidonoylglycerolu v hypothalamu, mozkové kůře a mozečku. Testovaná antidepresiva trazodon a venlafaxin významně zvyšují hladiny anandamidu a 2-arachidonoylglycerolu, zatímco mirtazapin významně snižuje pouze hladiny anandamidu.

Abstract:

Endocannabinoids have some pharmacological properties of antidepressants. The ones affect reuptake of monoamines, it can be considered as a potential therapeutic tool for treatment of mood and anxiety disorders. The endocannabinoid signaling is a key element in the regulation of mood and emotion. The damage of the endocannabinoid signaling leads to the development of depression. Increase of the endocannabinoid signaling could contribute to the same therapeutic effect, which current antidepressants have. The aim of this thesis is to contribute to expanding our knowledges about the modulation of the endocannabinoid signaling with using of the antidepressants and verify the effectiveness of the antidepressants which were used to the increase of the endocannabinoids, anandamide and 2-arachidonoylglycerol, in selected parts of the brain tissue. Mirtazapine, trazodone and venlafaxine were added to a standard laboratory diet to rats for 12 weeks. Samples of the brain tissue of the rats were divided into individual parts: the left hemisphere, right hemisphere, hypothalamus, cerebral cortex and cerebellum. The divided parts were homogenised, extracted and then measured by liquid chromatography with mass spectrometry. It was demonstrated that mirtazapine significantly reduces the concentration of anandamide in hypothalamus. While trazodone significantly increases the concentration of the anandamide in the right hemisphere, cerebral cortex and cerebellum. Elevated levels of the anandamide in the hypothalamus, cerebral cortex and cerebellum were reported after administration of venlafaxine. Concentration of the 2-arachidonoylglycerol was significantly increased in the left hemisphere, right hemisphere, hypothalamus, cerebral cortex and cerebellum after the administration of the trazodone. Venlafaxine significantly increased the concentration of the 2- arachidonoylglycerol in the hypothalamus, cerebral cortex and cerebellum. Tested antidepressants, the trazodone and venlafaxine significantly increase the levels of the anandamide and 2-arachidonoylglycerol. While mirtazapine significantly reduces only levels of the anandamide.
