Synthesis of new nucleoside analogs

Abstract:

V této bakalářské práci se věnujeme syntéze nových karbocyklických C-nukleosidových analogů. Prvá část práce je věnována uvedení do problematiky nukleosidových analogů, jejich výskytu v přírodě, struktuře, možným modifikacím a možnostem využití syntetických nukleosidových analogů. Druhá část práce je věnována popisu námi vyvinutých syntéz, vedoucích ke klíčovým meziproduktům a finálním látkám. Našim cílem bylo vyvinout účinnou a reprodukovatelnou syntézu vedoucí k 5´-amino intermediátu coby klíčové výchozí látky pro syntézu cílových látek. Z tohoto intermediátu jsme připravili čtyři finální látky, jejichž inhibiční aktivita vůči vybraným enzymům bude v budoucnu testována.

Abstract:

This bachelor thesis is focused on the synthesis of new carbocyclic C-nucleoside analogs. The first part of the thesis is dedicated to the introduction to the problematics of nucleoside analogs, their occurence in nature, structure, possible modifications, and the use of synthetic nucleoside analogs. The second part is dedicated to the description of the synthetic pathways we developed, leading to the key intermediate and final compounds. Our goal was to develop an effective and reproducible synthetic pathway leading to the 5´-amino intermediate as a key starting material for the synthesis of final compounds. From this intermediate, we prepared four final compounds, whose inhibitory activity towards selected enzymes will be tested in the future.
