Acute and chronic effects of sigma receptor ligands in mammalian myocardium

Abstract:

Sigma receptory byly poprvé popsány W.R. Martinem v roce 1976 v centrální nervové soustavě. Ani po téměř čtyřiceti letech trvajícího intenzivního výzkumu však doposud nebyly objasněny všechny otázky týkající se tohoto receptorového systému. Mezi ligandy sigma receptorů patří také mnohá antipsychotika schopná měnit elektrickou stabilitu myokardu. Již dlouho trvající zájem o srdeční sigma receptory, které byly studovány v naší laboratoři, nás přivedl na myšlenku studovat vedlejší účinky dvou typických ligandů sigma receptorů - haloperidolu a perfenazinu - na kardiovaskulární systém. Experimentální práce zahrnuté v těchto tezích byly zaměřené jak na akutní, tak chronické změny na elektrogramu u modelu izolovaného srdce perfundovaného dle Langendorffa. Práce byla doplněna o molekulárně – genetickou studii zaměřenou na expresi sigma 1 receptoru, IP3 receptoru typu 1 a 2 v jednotlivých částech srdce, v izolovaných kardiomyocytech a v buňkách PC12 po akutní expozici nebo chronické léčbě haloperidolem. Práce byla prováděna na dvou biomodelech – potkanovi a morčeti. Na základě současných literárních údajů a výsledků shrnutých v těchto tezích můžeme předpokládat, že srdeční sigma 1 receptory představují modulační systém transportu kalcia v srdeční buňce (spolu s IP3 receptory), a tak jemnou modulaci srdeční kontraktility a elektrických jevů.

Abstract:

Sigma receptors were originally referred by W.R. Martin in 1976 in central nervous system. Although since their discovery intense research has been aimed at understanding this receptor system, still numerous questions remain unanswered. Numerous antipsychotics are potent sigma receptor ligands susceptible to affect the electrical stability of myocardium. The long-lasting interest in cardiac sigma receptors which have been studied in our laboratory brought round the idea to study cardiovascular side effects of two typical representatives of sigma receptor ligands haloperidol and perphenazine. The experimental studies included in these theses were focused on both acute and chronic changes of electrogram recorded in isolated hearts perfused according to Langendorff. This part was completed with molecular genetic study focused on the expression of sigma 1 receptor, IP3 receptor of type 1 and 2 in the individual parts of the heart, in isolated rat cardiomyocytes and in PC12 cells after acute exposure or chronic treatment with haloperidol. Part of the abovementioned studies has been performed on two biomodels – rat and guinea pig. Based on the current literature knowledge and the results summarized in these theses, it may be hypothesized that cardiac sigma 1 receptors represent a modulatory system involved in cardiac calcium transportation (together with IP3 receptors) and thus fine modulation of cardiac contractility and it is also involved in modulation of membrane electrical events.
