The role of dendritic cells in diabetogeneic peptide presentation in patients with type 1 diabetes

Abstract:

Diabetes mellitus 1. typu je autoimunitní onemocnění charakterizované T-buněčnou destrukcí beta buněk Langerhansových ostrůvků pankreatu. Důležitou roli v jeho iniciaci a průběhu hrají dendritické buňky. DB jsou antigen prezentující buňky, které obecně vychytávají antigeny v lidském těle a pomocí receptorů na jejich povrchu je prezentují T-lymfocytům. U pacientů s DM1 je porušena funkce autotolerance, tudíž T-lymfocyty v závislosti na funkci dendritických buněk reagují na diabetogenní peptidy vyplavované do extracelulárního prostoru pankreatu. V této diplomové práci byla zkoumána role dendritických buněk při prezentaci diabetogenních peptidů u pacientů s DM1 a zdravých dárců. Cílem bylo pokusit se připravit tolerogenní dendritické buňky kultivací s IL-10 a TGF-β a porovnat jejich produkci cytokinů IL-12 a IL-10 po kultivaci s diabetogenními peptidy a autologními T-lymfocyty. Srovnávací skupinou tolerogenních DB byly buňky zdravých dárců a pacientů s DM1 ovlivněné aktivačním směrem kultivací s LPS a IFN.

Abstract:

T1D is an autoimmune disease caused by autoimmune reaction directed against autoantigens in the islets of Langerhans. This process leads to destruction of pancreatic beta cells and induces a whole-life dependence on exogenous doses of insulin. Dendritic cells play a very importatnt role in progress of this disease. DC are antigen presenting cells, in periphery capture and process antigens, express lyphocyte co-stimulatory molecules and secrete cytokines to initiate immune responses. They not only activate T-cells, they also tolerize T-cells to self-antigens and thus keep off the autoimmunity. In this study there was decribed the role of dendritic cells in presentation of diabetogeneic peptides in T1D patients and healthy controls. The aim was to try to prepare tolerogenic DC in vitro through cultivation in medium with IL-10 and TGF-β, and to compare a production of IL-12 and IL-10 by tolerogenic and activated (treated with LPS and IFN-γ) DC in presence of diabetogeneic peptides and T-cells.
