Analysis of MAPT gene in selected patients with neurodegenerative disease

Abstract:

Ve své diplomové práci se zabývám optimalizací metodiky pro vyšetření genu MAPT. Patogenní mutace v tomto genu souvisí s rozvojem frontotemporální lobární degenerace (FTLD) a jiných neurodegenerativních onemocnění jako je progresivní supranukleární obrna (PSP) nebo vzácněji Alzheimerova choroba (AD). V teoretické části se podrobněji věnuji statistickým údajům, klasifikaci a klinickému obrazu vybraných neurodegenerativních onemocnění, patofyziologickému podkladu jejich vzniku, možnosti léčby a metodám požívaným k jejich diagnostice. V praktické části práce se detailně zabývám genem MAPT a molekulární diagnostikou FTLD. Byly vyšetřeny všechny kódující exony genu MAPT-tzn. 2-15. Popisuji také metodiku celého pracovního procesu. Cílem této praktické části je vyloučení patogenních mutací v genu MAPT u vybraných pacientů s demencí.

Abstract:

In my diploma thesis I deal with the optimisation of diagnostics for testing of MAPT gene. Pathogenic mutations in this gene are linked to the development of frontotemporal lobar degeneration (FTLD), and other neurodegenerative diseases such as progressive supranuclear palsy (PSP), or rarely Alzheimer's disease (AD). The theoretical part includes in more detail description of statistics, classification and clinical conditions of selected neurodegenerative diseases, pathophysiological basis, treatment options and methods used for their diagnostics. In practical part of the thesis I describe MAPT gene and the molecular diagnostics in more detail. All coding exons of the MAPT gene-ie. 2 to 15-were examined. I also describe the methodology of the entire laboratory work process. The aim of the practical part is to screen out pathogenic mutations in MAPT gene in a selected group of patients with dementia.
