Intraspecific variability in Platyhelminthes: review of available techniques and possible application of RAD sequencing

Anotácia:

Tato bakalářská práce je literární rešerší zabývající se vnitrodruhovou variabilitou obligátně parazitických Platyhelminthes a technikami, běžně využívanými k hodnocení této variability. V práci je kladen důraz na zhodnocení možnosti využití RAD sekvenování. První část práce je věnována životním strategiím jednotlivých skupin parazitických Platyhelminthes (Trematoda, Cestoda, Monogenea) a zaměřena na jejich morfologickou a molekulární charakteristiku. Druhá část se zabývá typy parazitických populací a jejich charakteristikou. Třetí část shrnuje metody hodnocení vnitrodruhové variability, spolu s případovými studiemi zabývajícími se danou problematikou. Tato část popisuje morfologické a genetické metody využívající restrikčních enzymů, a prezentuje dosavadní případové studie kombinující morfologické a molekulární přístupy u třídy Monogenea. V poslední části práce je představen princip RAD sekvenování, možnosti jeho využití v parazitologii a návrh protokolu vhodného pro studium populační genetiky třídy Monogenea.

Abstract:

This bachelor's thesis is a review, which focuses on an intraspecific variability of parasitic members of Platyhelminthes and techniques, which are commonly used to document this variability. The main aim is the evaluation of RAD sequencing as a new method in this field. First part of the thesis describes the life style of different groups of parasitic Platyhelminthes (Trematoda, Cestoda, Monogenea) concentrating on morphology and molecular characterisation. Second part refers to parasitic populations and their characteristics. Third part summarizes methods involved in the evaluation of intraspecific variability, together with case studies. This part is divided into sections, where strictly morphological or genetic restriction-based methods are documented and presents case studies, which have combine morphology and genetics in research of monogeneans so far. Last part presents RAD sequencing by mean to evaluate its use in the field of Parasitology. As a result, protocol suitable for populations genetics in monogeneans is proposed.
