Influence of plasmachemical modifications of polymers on cytokinetic parameters of cells

Abstract:

Již řadu let je snahou tkáňového inženýrství vyvinout takové tkáňové náhrady, které by byly tělu přirozené, samy se vstřebávaly a byly nosičem pro tělu vlastní buňky. Na popularitě nabírají syntetické polymery modifikované relativně jednoduchými, po ekonomické i ekologické stránce vhodnými, plazmochemickými procesy, díky kterým se stávají pro buňky vhodným nosičem. Tato práce se zaměřuje na polykaprolaktonové nanovlákenné membrány plazmochemicky pokryté buď netoxickým cyklopropylaminem (CPA), který vede k vytvoření vrstev bohaté na pozitivně nabité aminové skupiny, nebo kopolymerizací kyseliny maleinové s acetylenem za vzniku negativně nabitých karboxylovaných vrstev (BioMA). Cílem této diplomové práce bylo zjistit účinek takto funkcionalizovaných povrchů na lidské buňky, konkrétněji keratinocyty linie HaCaT. Předmětem výzkumu byl vliv na proliferaci, diferenciaci, adhezi a buněčnou smrt těchto kožních buněk. Pomocí imunocytochemické detekce strukturních proteinů bylo testováno, zda buňky nepodléhají zásadním změnám morfologie či nepodléhají přestavbě spojené s některým z cytokinetických procesů. Na rozdíl od předchozích experimentů prováděných na myoblastové linii C2C12, HaCaT buňky rostly stejně rychle na membránách nefunkcionalizovaných i funkcionalizovaných. Je nutné také zmínit, že jejich míra adhezivity k povrchu byla vyšší na upravených površích. Tento fakt se odráží také ve změnách v expresi a lokalizaci některých strukturních proteinů (vimentin, tubulin βIII, fibronektin, E kadherin etc.) a zahrnuje také aktivaci  kateninové dráhy.

Abstract:

For many years, there has been an effort of tissue engineering to develop tissue replacements that would be natural to the body, gradually absorbed by the body and be suitable for carrying new cells. Synthetic polymers are gaining popularity especially when they are modified by relatively simple, economically and ecologically suitable plasmachemical processes, making them a suitable carrier for the cells. This diploma thesis focuses on polycaprolactone nanofibrous membranes plasmachemically treated with either non-toxic cyclopropylamine (CPA), which leads to the formation of layers rich in positively charged amine groups or by copolymerization of maleic acid with acetylene to form negatively charged carboxylated layers (BioMA). The aim of this thesis was to test whether these functionalized layers affect human cells, more specifically the keratinocytes of HaCaT line. The subject of the study was the influence on the, proliferation, death, adhesion and differentiation of these skin cells. Using the immunocytochemical detection of structural proteins, it was tested whether the cells were subjected to major morphological alterations or to rebuilding associated with some of the cytokinetic processes. Unlike the previous experiments performed on the C2C12 myoblast line, HaCaT cells grew equally fast on nonfunctional and functionalized membranes. It should also be noted that their adhesion was higher on the treated surfaces. This corresponds to changes in the expression and localization of some structural proteins (vimentin, tubulin βIII, fibronectin, E cadherin etc.) and there is active catenin pathway involved.
