Regulation of expression and function of miR-34a tumor suppresor in the cells with inactive form of TP53

Abstract:

MicroRNAs belong to small non-coding RNA molecules and are extensively studied in relation to carcinogenesis. One of the most studied microRNAs is miR-34a, an important tumour supressor, which is included in regulation of many cellular processes, such as differentiation, proliferation, and apoptosis. Anti-tumour functions of miR-34a are realized in cooperation with another tumour supressor – TP53. In many types of cancer TP53 is mutated or inactive, and the functions of miR-34a in such cells are not well understood. Successful therapeutic application with defective function of p53 in cancer requires thorough understanding of the mechanisms which control miR-34a. The goal of this thesis was to clarify the function of miR-34a in cells with dysfunctional TP53 signalization and to describe other mechanisms by which it can be regulated. The first part of the work demonstrates that in such cells miR-34a regulates its validated target genes CD44, CD24 and MDMX. The second part proves an inhibiting effect of p63 on miR-34a in prostate cells with dysfunctional TP53. These results contributed to the understanding of function of miRNA in the context of cells with disrupted p53 function.

Abstract:

MicroRNA patří mezi krátké nekódující RNA molekuly a jsou intezivně studované ve vztahu ke karcinogeneze. Jednou z nejlépe prostudovaných miRNA je miR-34a. Je to důležitý tumorový supresor, který se zapojuje do regulace mnohých buňkových procesů včetně diferenciace, proliferace a apoptózy. Protinádorové funkce miR-34a jsou důsledkem spolupráce s dalším tumorovým supresorem - TP53. Ten je v mnohých typech nádorů mutovaný či neaktivní a funkce miR-34a jsou v těchto buňkách málo známé. Předpokladem úspěšné terapeutické aplikace při narušené funkci p53 v rakovinových onemocnění je důkladné pochopení mechanizmů, které řídí miR-34a. Cílem této práce proto bylo objasnit funkci miR-34a v buňkách s nefunkční signalizací TP53 a popsat další mechanismy, kterými může být regulovaná. V první části práce bylo ukázáno, že miR-34a reguluje v těchto buňkách své validované cílové geny CD44, CD24 a MDMX. Ve druhé části práce byl prokázán inhibiční vliv p63 na miR-34a v prostatických buňkách s nefunkční TP53. Těmito výsledky jsme přispěli k porozumění funkce miRNA v kontextu buněk s narušenou funkcí p53.
