Detoxification of pollutants by plants,affecting of model organisms by toxic compounds-Utilizing for phytoremediation technologies

Abstract:

Stále vyšší obsah toxických látek v životním prostředí je spojen především s průmyslovou a zemědělskou činností člověka (spalování fosilních paliv, používání umělých hnojiv, pesticidů, detergentů atd.). Mezi jedny z velmi nebezpečných toxických látek, kterou jsou schopny akumulovat v různých složkách životního prostředí, patří perzistentní organické polutanty (POPs). Do této skupiny látek jsou zahrnuty dioxiny, furany, ale také organochlorované pesticidy a řada dalších látek. Nedávno byly objeveny rostlinné druhy, které jsou schopny organické polutanty akumulovat bez viditelné deprese a zpomalení růstu. Detailní mechanismus a působení na různé rostlinné enzymy není zcela znám. Proto jsme se v této práci zaměřili na studium vlivu různých POPs na růst rostlin kukuřice a obsah thiolových sloučenin (cysteinu Cys, redukovaného GSH a oxidovaného GSSG glutathionu, a fytochelatinu PC2). Nejprve bylo nezbytné optimalizovat techniku vhodnou pro citlivé a simultánní stanovení thiolových látek. Pro tyto účely jsme zvolili techniku vysoce účinné kapalinové chromatografie s elektrochemickou detekcí. Nejvhodnější HPLC-ED parametry pro citlivou a simultánní detekci studovaných thiolů byly gradientová mobilní fáze skládající se z 80 mM trifluoroctové kyseliny a methanolu, průtoku mobilní fáze 0.8 ml min-1, teploty kolony a detektoru 25 stupňů Celsia, a elektrodového potenciálu 900 mV. Za těchto podmínek jsme byly schopni stanovit desítky femtomolů (3 S/N) analyzovaných thiolů na nástřik (5 ul). V další části práce jsme se zabývali studiem vlastního vlivu POPs na rostliny kukuřice. V našich experimentech byly rostliny kukuřice seté (Zea mays) vystaveny působení 0, 0.1, 0.3, 0.5, 0.7 a 1 nM směsi POPs po dobu čtrnácti dnů za kontrolovaných podmínek. Prvním studovaným parametrem vlivu rozdílné koncentrace chlorovaných uhlovodíků na rostliny kukuřice (odrůda Gila) byly základní růstové charakteristiky. V průběhu experimentu jsme zjistili, že POPs velmi intenzivně stimulují růst kukuřice. V dalších experimentech byl studován obsah Cys, GSH, GSSG a PC2 při jednotlivých studovaných koncentrací POPs v nadzemních částech rostliny a v kořenech. V kořenech byly obsahy jednotlivých thiolů mírně zvýšené v porovnání s nadzemními částmi rostlin, což zřejmě souvisí s procesy, které probíhají přímo v kořenech. Dále jsme se zaměřili na sledování vlivu POPs na klíčení kukuřice a celkový obsah thiolů. Zjistili jsme, že nejvyšší obsah thiolů byl pozorován u aplikované koncentrace POPs 0.3 nM, přičemž tato koncentrace nejvíce stimulovala klíčení.

Abstract:

Increasing amounts of toxic compounds in environment is associated mostly with human industrial and agricultural activities (products of combustion of fossil fuels, using of artificial fertilizers, pesticides, surfactants, etc.). Persistent organic pollutants (POPs) belong to the most toxic and dangerous compounds due to their ability to tend to accumulate in different parts of environment. Dioxins, furans, organochlorinated pesticides and many others substances are included to POPs. Recently, plants species, which can accumulate organic pollutants without decreasing growth rate and depression, have been investigated. The mechanism of this phenomenon and its influence on plant enzymes are not clear yet. Thus, we aimed on investigation of influence of POPs on growth and thiols content (cysteine Cys, reduced GSH and oxidized GSSG glutathione, and phytochelatin PC2) in maize plants. Primarily, optimising of a suitable technique for sensitive and simultaneous determination of the thiols was need. We choose high performance liquid chromatography with electrochemical detection for these purposes. These thiol compounds were well separated and detected at most suitable HPLC-ED conditions as follows - gradient mobile phase: 80 mM trifluoroacetic acid and methanol, flow rate 0.8 ml min-1, column and detector temperature 25 Celsius degrees and electrode potential 900 mV. We were able to determine tens of femtomoles (3 S/N) of the thiols per injection (5 ul). After that, we focused on investigation of the influence of POPs on maize plants. Maize plants (Zea mays) were treated by POPs (0, 0.1, 0.3, 0.5, 0.7 and 1 nM) for fourteen days under controlled conditions. Here, we studied primarily the basic growth parameters of the treated plants. We found out that POPs stimulated intensively growth of maize plants in comparison with control. Consequently, we analysed the roots and aboveground parts of the treated plants by HPLC-ED technique and quantified Cys, GSH, GSSG and PC2. The thiols content in roots were slightly higher in comparison with aboveground parts. This phenomenon probably relates with detoxification processes taking place in the roots. After that, we focused on investigation of influence of POPs on germination of maize kernels and thiols content. We found out that the highest thiols content were determined in maize kernels treated by POPs (0.3 nM), whereas this POPs concentration stimulates mostly germination.
