Determination and diagnostic significance of essential fatty acids in diabetic patients type 2

Abstract:

V rámci této diplomové práce byl zkoumán obsah esenciálních mastných kyselin v plazmě diabetiků a kontrolní skupiny. Byla vytvořena jejich závislost na onemocnění diabetes mellitus 2. typu, které je dáno hladinou glykovaného hemoglobinu (HbA1c). Cílem této práce bylo charakterizovat markery z řad polynenasycených mastných kyselin, které by mohly přispět k včasné diagnostice diabetu. Vzorky plazmy zdravých dárců a diabetiků 2. typu byly analyzovány dvěma chromatografickými metodami. Nejdříve byla provedena tenkovrstvá chromatografie, na jejíž základě byly plazmatické lipidy rozděleny do pěti lipidových tříd. Následně byla provedena derivatizace na methylestery mastných kyselin a plynovou chromatografií byla stanovena koncentrace jednotlivých mastných kyselin. Tímto způsobem bylo zpracováno 32 vzorků plazmy, které byly rozděleny do dvou skupin podle hodnot glykovaného hemoglobinu (HbA1c) na diabetiky 2. typu a kontrolní skupinu (zdravé dárce). U sledovaných diabetiků byla zjištěna snížená aktivita 5-desaturázy, což charakterizuje orgány postižené diabetem v počátku jejich postižení. Naproti tomu byla u diabetiků zjištěna zvýšená aktivita 6-desaturázy, která souvisí s progresí metabolického syndromu. Statistickou analýzou byly odhaleny čtyři polynenasycené mastné kyseliny, které by se daly použít pro včasnou diagnostiku onemocnění diabetes mellitus 2. typu.

Abstract:

In this diploma thesis the content of essential fatty acids in the plasma of diabetics and control group was examined. There were evaluated their dependence on type 2 diabetes mellitus, which is determined by the level of glycated hemoglobin (HbA1c). The aim of this work was to characterize markers from polyunsaturated fatty acids that could contribute to the early diagnosis of diabetes. Plasma samples of healthy donors and type 2 diabetics were analysed by two chromatographic methods. At first, thin layer chromatography was performed and the plasma lipids were divided into five lipid classes. The derivatization of fatty acid into corresponding methyl esters was carried out and the concentration of individual fatty acids was determined by gas chromatography. Thirty two plasma samples were processed in this way. According to the values of glycated hemoglobin (HbA1c), samples were divided into two groups: diabetics of 2type and control group (healthy donors). Reduced activity of 5-desaturase was observed within diabetics group. This is characteristic for organs which are affected by diabetes at the beginning of their disability. On the other hand, increased 6-desaturase activity has been found in diabetics, which is related to the progression of metabolic syndrome. Statistical analysis revealed four polyunsaturated fatty acids that could be used for the early diagnosis of type 2 diabetes mellitus.
