Effect Of Sample Preparation On The Electrochemical Determination Of Ergosterol Using Selected Electrodes

Abstract:

Cílem práce bylo elektrochemické stanovení ergosterolu a zavedení vhodné úpravy vzorku pro měření pomocí extrakce stanovované látky. V teoretické části jsou shrnuty obecné informace o ergosterolu, jeho funkce v souvislosti s vitamínem D a korelace jeho obsahu s přítomností některých mykotoxinů. V dalších kapitolách jsou popsány způsoby extrakce ergosterolu ze vzorku, z nichž vybrané byly následně využity v experimentální části práce. Dále jsou popsány vybrané typy elektrod, které byly použity pro detekci ergosterolu a porovnány. Praktická část je věnována optimalizaci některých podmínek elektrochemické detekce pro použité elektrody porovnání vybraných extrakčních postupů. Optimalizace a stanovení výtěžnosti extrakcí bylo provedeno s pomocí standardu ergosterolu. Extrakce s nejvyšší výtěžností byla použita pro stanovení reálných vzorků čistých plísní narostlých na médiu a uměle kontaminovaných vzorků potravin. V závěrečné části práce jsou popsány a diskutovány získané výsledky.

Abstract:

The goal of the work was the electrochemical determination of ergosterol and the introduction of a suitable preparation of the sample for measurement using the extraction of the determined substance. The theoretical part summarizes general information about ergosterol, its function in connection with vitamin D and the correlation of its content with the presence of some mycotoxins. In other chapters there are descriptions of methods of extracting ergosterol from the sample, some of which were subsequently used in the experimental part of the work. In next chapters there are descriptions and comparisons of selected types of electrodes that were used for the detection of ergosterol. The practical part is devoted to the optimization of some conditions of electrochemical detection for used electrodes and the comparison of selected extraction procedures. Optimization and determination of the extraction yield was carried out with the help of the ergosterol standard. The extraction with the highest yield was used for the determination of real samples of pure fungi grown on media and artificially contaminated food samples. In the final part of the thesis, the obtained results are described and discussed.
