Synthesis of pyrazolo[1,5-a]pyrimidine-based inhibitors of CDK kinases

Abstract:

Nádorová onemocnění patří k nejzávažnějším zdravotním problémům 21. století. Mnoho výzkumných zařízení se pokouší najít lék, který by pomohl v boji proti rakovině. Zásadním problémem je ovšem velké množství druhů rakovinových nádorů, jenž mohou vznikat různými způsoby a najít jeden lék na všechny typy tohoto onemocnění je téměř nemožné. Tato práce se zaměřuje na inhibici cyklin-dependentních kináz (CDK), jenž jsou látky, které mají velice významné úlohy v lidském těle. Dojde-li k chybě v DNA, může se tato chyba přenést na tyto kinázy při jejich syntéze a zapříčinit jejich nadměrnou aktivitu. Ta může způsobit nekontrolovatelné množení buněk a tvorbu nádorů. Zaměřili jsme se na syntézu 4 inhibitorů CDK, jejichž základní struktura je založena na pyrazolo[1,5-a]pyrimidinu. První z nich je látka SCH 727965, která se nachází v 2. fázi klinického testování a ukázala velice dobré výsledky v boji proti rakovině vaječníku. Další tři látky jsou analogy známého inhibitoru olomoucinu II, jenž byly syntetizovány pro porovnání jejich aktivit.

Abstract:

Cancer is one of the most serious health challenges of the 21st century. Many research institutes are trying to find a drug that would help in the fight against cancer. The fundamental problem is that exists many types of cancers tumors which can develope in different ways and find a cure for all types of this disease is almost impossible. This work focuses on the inhibition of cyklin-dependent kinases (CDKs), which are substances that have very important roles in the human body. If an error occurs in the DNA, this mistake may be transferred to the kinases in their synthesis and the cause of their excessive activity. This can cause uncontolled cell proliferation and tumor formation. We focused on the synthesis of four inhibitors of CDKs, whose basic structure is based on pyrazolo[1,5-a]pyrimidine. The first is the substance SCH 727965, which is located in the 2nd phase of clinical testing and has shown very good results in the fight against ovarian cancer. Three other agents are analogues of known inhibitors olomoucine II, which were synthesized to compare their activities.
