Resistance Evaluation of Genetic Resources, Reference Clones and Progenies of Hybrid Combinations to Apple Scab

Abstract:

Rezistentní šlechtění jabloní vůči strupovitosti způsobené houbovým patogenem Venturia inaequalis (Cooce) G. Winter, je v současnosti jedna z nejdůležitějších strategií ochrany vůči této chorobě. Pěstování odrůd s účinnou polní rezistencí má pozitivní dopad nejen v rentabilitě produkce jablek, ale je kladně vnímána i z pohledu snížení množství reziduí fungicidů v plodech i v celém životním prostředí. Z hlediska šlechtitelského je nejjednodušší se zaměřit na monogenně podmíněnou rezistenci. Na základě vhodně sestaveného šlechtitelského plánu, který bývá mnohdy doplněn molekulárními markery, lze relativně rychle vyšlechtit rezistentní odrůdu. Velkým problémem však je trvanlivost této rasově specifické rezistence. Nejčastěji využívaným majorgenem rezistence jabloní vůči strupovitosti je gen Rvi6, pocházející z botanického druhu Malus floribunda. Masové rozšíření odrůd s tímto genem v produkčních sadech vyvolalo selekční tlak v populacích V. inaequalis. Došlo k selekci agresivních ras, které tuto rezistenci překonaly. Diplomová práce je proto zaměřena na hodnocení variability odolnosti diferenciačních klonů jabloně v českých i evropských agroekologických podmínkách v letech 2014 ̶ 2019. Statistická analýza (GLM – ANOVA) prokázala, že statisticky významnými zdroji variability (α = 0,05) je genotyp diferenciačního klonu, ročník a lokalita. Podrobnější analýza prokázala pomocí Tukeyova HSD testu (α = 0,05) existenci homogenních skupin. Například v analyzovaných datech evropského projektu VINQUEST byla odlišena francouzská lokalita Angers s výrazně nižším stupněm odolnosti způsobeným výskytem agresivních ras pocházejících z nekulturních rostlin. V podmínkách České republiky byla zjištěna vysoká korelace (r2 > 0,65) mezi odolností klonů a průběhem počasí. Byla zpracována rovněž robustní statistická analýza zaměřená na segregaci imunních, hypersenzitivních a celkově odolných potomků v hybridních generacích vzniklých ve VŠÚO Holovousy s.r.o. v letech 2004 ̶ 2007. Analýza byla zaměřena na rodičovské kombinace, které se z hlediska výskytu imunních, hypersenzitivních a odolných potomků nacházely v prvním a druhém decilu. Pomocí χ2-testu byly zjištěny významné rozdíly (α = 0,05) štěpných poměrů mezi hodnocenými ročníky. Regresní a korelační analýza potvrdila trend (r2 > 0,87) v poklesu výskytu odolných potomků. Dále byl navržen algoritmus výpočtu predikčních koeficientů vhodných pro identifikaci rodičů schopných poskytovat odolné potomky. Výsledky predikční analýzy ukázaly, že v roce 2007 byly nejlépe hodnoceny genotypy odvozené od šlechtitelského materiálu OR-45T-132, nesoucího rezistentní gen Rvi5. Tento výsledek umožnil formulovat závěr, že inokulační suspenze byla v roce 2007 připravena z ras V. inaequalis překonávajících rezistenci založenou na bázi genu Rvi6. Kombinace kodominantního markeru AL07 (Tartarini et al., 1999) a dominantního markeru VfA-VfD (Vávra et al., 2015b), byla použita pro identifikaci alel genu Rvi6. Metoda byla validována u souboru 96 odrůd se známou odolností vůči strupovitosti. Segregace markerů byla následně hodnocena u dvou hybridních potomstev s odlišným genetickým založením rodičů. U obou markerů byla pomocí χ2-testu potvrzena mendelistická segregace (α = 0,05). Přístup moravských ovocnářských podniků k pěstování rezistentních odrůd jabloní byl hodnocen na základě dotazníkového šetření. Všichni pěstitelé potvrdili prolomení rezistence řízené genem Rvi6. Někteří pěstitelé uvedli u těchto odrůd možnost méně intenzivní fungicidní ochrany. Nejkladněji byly hodnoceny dvě české rezistentní odrůdy ̶ Topaz a Rubinola. Abstract:

The apple breeding programs focused on the resistance against the apple scab, which is caused by the fungal pathogen Venturia inaequalis (Cooce) G. Winter, are currently one of the most important strategies for protection against this disease. The cultivation of varieties with effective field resistance has a positive impact not only in the profitability of apple production, but it is also positively perceived in terms of reducing the amount of fungicide residues in fruits and in the whole environment. From the breeding point of view, the easiest strategy is focusing on monogenic resistance. On the basis of a suitably compiled breeding plan, which is often supported by molecular markers, resistant varieties can be bred relatively quickly. However, the biggest problem is the durability of this racially specific resistance. The most frequently used majorgen of apple resistance to scab is the Rvi6 gene which is derived from the botanical species Malus floribunda. Unfortunately, a mass distribution of varieties with this gene into production orchards caused a selection pressure in V. inaequalis populations. A selection among races of V. inaequalis has occurred. Aggressive races were selected and these races were able to overcome this resistance. This diploma thesis is therefore focused on the evaluation of the variability of resistance of apple differentiation clones in Czech and European agroecological conditions in the years 2014 ̶ 2019. Statistical analysis (GLM ̶ ANOVA) proved that the genotype of the differentiation clone, year and locality are statistically significant sources of variability (α = 0.05). More detailed analysis showed the existence of homogeneous groups using Tukey's HSD test (α = 0.05). For example, in the analysed data of the European project VINQUEST, the French locality of Angers with a significantly lower degree of resistance caused by the occurrence of aggressive races originating from non-cultivated plants, was distinguished. In the conditions of the Czech Republic, a high correlation (r2 > 0.65) was found between the resistance of clones and the course of the weather. A robust statistical analysis focused on the segregation of immune, hypersensitive and generally resistant offspring in hybrid generations was created at Research and Breeding Institute of Pomology Holovousy Ltd. in 2004 ̶ 2009. The analysis was focused on parental combinations, which were in the first and second decile in terms of the occurrence of immune, hypersensitivity and resistant offspring. Using the χ2-test, significant differences (α = 0.05) of segregation ratios between the evaluated years were found. The regression and correlation analysis confirmed a decreasing trend (r2 > 0.87) in the incidence of resistant individuals in the offspring. Furthermore, there was proposed an algorithm for calculating of predictive coefficients suitable for identifying parents capable of providing resistant offspring. The results of the prediction analysis showed that genotypes derived from the breeding material OR-45T-132 carrying the Rvi5 resistance gene were evaluated as the best ones in 2007. This result enabled to formulate the conclusion that the inoculum suspension, which was used in 2007, was prepared from races of V. inaequalis which overcome the resistance based on the Rvi6 gene. The combination of the codominant marker AL07 (Tartarini et al, 1999) and the dominant marker VfA-VfD (Vávra et al., 2015b) was used to identify alleles of the Rvi6 gene. The method was validated for a set of 96 varieties with known scab resistance. The segregation of markers was subsequently evaluated in two hybrid offspring with different parental genetic background. Mendelian segregation (α = 0.05) was confirmed for both markers using the χ2-test. wn of Rvi6-driven resistance. Some growers have indicated the possibility of less intensive fungicidal protection for these varieties. Two Czech resistant varieties - Topaz and Rubinola - were evaluated the most positively. On the contrary, below-average fruit quality was reported for foreign varieties Prima and Florina.
