Sublethal effects and biomarkers of cyanobacterial ecotoxicity

Abstract:

Sinice neboli cyanobakterie jsou prokaryotické organismy známé svou schopností vytvářet vodní květ. Ten vzniká v eutrofizovaných vodách a je celosvětovým problémem, neboť znesnadňuje využití vodních nádrží a negativně ovlivňuje ostatní vodní organismy i celý ekosystém. Jedním z mechanismů tohoto negativního působení je produkce sinicových toxinů – cyanotoxinů. Sinice jsou v různých částech světa spojovány s hromadnými úhyny vodních živočichů a jejich negativní účinky byly pozorovány také v experimentálních studiích. Tyto studie ukázaly, že sinice a jejich toxiny mají prokazatelně schopnost po dlouhodobém působení vyvolávat změny biochemických, hematologických, fyziologických a morfologických parametrů. Změny těchto parametrů jsou užívány jako tzv. časné varovné signály neboli biomarkery ekotoxicity sinic. Tato práce se zabývá působením sinic na biochemické biomarkery ve studiích s křepelkou japonskou (Coturnix japonica) a potkanem (Rattus norvegicus var.alba). Studie se zaměřily na dlouhodobé působení sinic na ptáky v koexpozicích s jinými stresory prostředí a vliv sinic na sekundární konzumenty – savce. Křepelky byly ve studii vystaveny koexpozici biomasy sinic (s převládajícím druhem Microcystis), olova ve formě broků a patogenních mikroorganismů ve formě vakcíny. Potkani byli krmeni potravou s obsahem rybího masa a sinicových toxinů microcystinů. Byl sledován vliv na biochemické parametry oxidativního stresu a detoxikace. V rámci této práce byla také provedena optimalizace metod stanovení nových biomarkerů oxidativního stresu – enzymatické aktivity superoxid dismutázy (SOD) a katalázy (CAT). Ve studii s křepelkou byl v tkáni jater a srdce zaznamenán signifikantní nárůst hladiny redukovaného glutathionu (GSH), enzymatické aktivity glutathion-S-transferázy (GST), glutathion peroxidázy (GPx), glutathion reduktázy (GR), superoxid dismutázy (SOD) a změny míry lipidní peroxidace v závislosti na zvolené expozici. Ve studii s potkany došlo v jaterní tkáni k signifikantnímu nárůstu enzymatické aktivity glutathion-S-transferázy (GST), glutathion reduktázy (GR) a míry lipidní peroxidace. Z výsledků práce vyplývá, že sinice a jejich toxiny vyvolaly u sledovaných organismů oxidativní stres a zvýšenou míru detoxikace.

Abstract:

Blue-green algae or cyanobacteria are prokaryotic organisms that are known for their ability to form water blooms. Blooms rise in eutrophic water bodies and they cause worldwide problems with use of these waterbodies and can negatively influence aquatic ecosystem and organisms. The production of cyanobacterial toxins – cyanotoxins is one of the mechanisms of their ecotoxicity. Cyanobacteria have been connected with mass mortalities of aquatic animals in various parts of the world. Their negative effects have been observed in experimental studies as well. These studies showed that cyanobacteria and their toxins are able to cause changes of biochemical, hematological, physiological and morphological parameters. The changes of these parameters are used as early-warning signals or biomarkers of cyanobacterial ecotoxicity. This work focused on effects of cyanobacteria on biochemical biomarkers in studies with Japanese quail (Coturnix japonica) and rat (Rattus norvegicus var. alba). The studies examined long-term effects of cyanobacteria on the birds in co-exposure with other environmental stressors and on effects of cyanobacteria on secondary consumers – mammals. Japanese quails were co exposed to cyanobacterial water bloom (dominated by Microcystis), lead shots and pathogenic microorganisms in vaccine. Rats were fed by feed with fish meat and cyanobacterial toxins microcystins. Effects on biochemical parameters of oxidative stress and detoxification were studied. A part of this work was also the optimalization of assessment methods of new biomarkers of oxidative stress – enzymatic activities of superoxide dismutase (SOD) and catalase (CAT). In study with Japanese quail significant increase of glutathione level (GSH), the activity of glutathione-S-transferase (GST), glutathione peroxidase (GPx), glutathione reductase (GR), superoxide dismutase (SOD) and changes of lipid peroxidation were observed in liver and heart in dependence of the type of exposure. In study with rats significant increase of the activity of glutathione-S-transferase (GST), glutathione reductase (GR) and changes of lipid peroxidation were observed in liver. The results of this work show that cyanobacteria and their toxins induced oxidative stress and increase of detoxification in tested organisms.
